Fast Protein Liquid Chromatography

Fast protein liquid chromatography (FPLC) is a liquid chromatography method used to separate, purify, and analyze proteins under controlled conditions. The system pumps buffered aqueous solutions through a column containing a stationary phase, while proteins separate according to properties such as size, charge, or affinity; detectors monitor eluted components as the mobile-phase composition changes. Common modes include size-exclusion, ion-exchange, and affinity chromatography, which can be selected for a target protein and sample characteristics. In chemistry and biochemistry, FPLC supports protein purification, isolation of complexes, and preparation of samples for structural, enzymatic, and functional studies.

High-performance liquid chromatography, or HPLC, is an analytical technique that separates liquid samples under high pressures. An HPLC instrument consists of glass bottles for storing solvents called mobile phase reservoirs. HPLC-grade solvents are used to maintain high purity, and the dissolved gases are removed using a degasser, such as a vacuum pumping system or sparging with helium. The solvents are then pumped into the analytical column using a screw-driven syringe or reciprocating pumps.
