Barley Leaf Sheath

The barley leaf sheath is the basal portion of a leaf that wraps around the stem, protecting developing tissues and helping position the expanding leaf blade. During development, cells in the leaf primordium proliferate, elongate, and differentiate into sheath tissues as vascular strands form and the sheath extends around successive stem regions. Its growth reflects coordinated regulation of cell division, cell expansion, tissue patterning, and environmental or hormonal signals. Studying the barley leaf sheath helps developmental biologists understand monocot leaf formation, stem-leaf architecture, and the cellular basis of differences in plant growth, structure, and productivity.

Many developmentally important genes have cell- or tissue-specific expression patterns. This paper describes LM RNA-seq experiments to identify genes that are differentially expressed at the maize leaf blade-sheath boundary and in lg1-R mutants compared to wild-type. The experimental considerations discussed here apply to transcriptomic analyses of other developmental phenomena.

Relating Stomatal Conductance to Leaf Functional Traits

Unraveling how physiology and morphology are linked allows a deeper understanding of mechanistic functioning of plant leaves. We present both a procedure to derive parameters of stomatal regulation from stomatal conductance measurements and correlations with traditional functional leaf traits.

We have developed a simple and reproducible protocol to access stomatal response to live bacteria. This method minimizes wounding and manipulation of the leaf as compared to the use of epidermal peels reported previously.

A method for metabolomic analysis of barley is presented. The method entails fractionation and derivatization of metabolites and analysis thereof by gas chromatography/mass spectrometry (GC/MS). Metabolomic analysis can be used to determine the effect of intercropping and drought stress on the grain.
