Embryo Orientation

Embryo orientation is the establishment and maintenance of an embryo’s spatial axes, such as anterior-posterior, dorsal-ventral, and left-right, which enables cells to acquire position-specific identities during development. It arises through coordinated polarity cues, localized signaling centers, morphogen gradients, and directed cell movements that translate molecular asymmetries into organized tissue geometry. In developmental biology, analyzing embryo orientation helps explain how body plans form, how organs acquire consistent placement, and how disruptions produce patterning defects. Imaging, lineage tracing, and genetic manipulation of orientation pathways provide experimental frameworks for linking early embryonic signals to later morphology and comparing conserved mechanisms across species.

The human heart, despite its modest size and weight, is an organ of remarkable strength and endurance. Roughly the size of a fist, the heart weighs between 250 and 350 grams and is nestled within the mediastinum, the medial cavity of the thorax. It extends obliquely for about 12 to 14 cm, resting on the superior surface of the diaphragm. The heart is positioned anterior to the vertebral column and posterior to the sternum, with two-thirds of its mass lying to the left of the midsternal line.

In-vivo Centrifugation of Drosophila Embryos

We describe a method to separate organelles by density in living Drosophila embryos. Embryos are embedded in agar and centrifuged. This technique yields reproducible separation of major organelles along the anterior-posterior embryo axis. This method facilitates colocalization experiments and yields organelle fractions for biochemical analysis and transplantation experiments.
