Fluorescence Imaging Analysis

Fluorescence imaging analysis is the quantitative interpretation of images produced by fluorescent labels, converting patterns of emitted light into information about cells, molecules, and tissues. Fluorophores absorb excitation light and emit longer-wavelength light, while image-analysis workflows measure signal intensity, location, shape, and changes over time after background correction and segmentation. In developmental biology, these measurements help researchers track gene expression, cell division, migration, differentiation, and tissue morphogenesis in fixed or living specimens. By linking fluorescence patterns with developmental stages and cellular behaviors, the approach supports precise comparisons across experiments and reveals how molecular signals guide the formation of complex organisms.

We present an open source high content analysis (HCA) instrument utilizing automated fluorescence lifetime imaging (FLIM) for assaying protein interactions using Förster resonance energy transfer (FRET) based readouts. Data acquisition for this openFLIM-HCA instrument is controlled by software written in µManager and data analysis is undertaken in FLIMfit.

Here we present a reliable method to monitor the incorporation of nanoparticles into a polymer host matrix via swell encapsulation. We show that the surface concentration of cadmium selenide quantum dots can be accurately visualized through cross-sectional fluorescence imaging.
