Granulosa Cell Removal

Granulosa cell removal is a laboratory procedure that separates ovarian oocytes from surrounding granulosa and cumulus cells, enabling direct study of oocyte biology and early development. Typically, enzymatic dissociation with hyaluronidase weakens the extracellular matrix, followed by gentle mechanical pipetting to release the denuded oocyte while preserving its structure. In developmental biology, this preparation supports assessments of oocyte maturation, fertilization, activation, and preimplantation embryo development under controlled conditions. It also helps researchers distinguish oocyte-intrinsic effects from signals provided by surrounding somatic cells, although excessive exposure to enzymes or mechanical force can compromise oocyte quality and developmental competence.
