Bacterial Biomarker Detection

Bacterial biomarker detection is the identification and measurement of molecular signatures associated with bacterial cells, such as nucleic acids, proteins, or metabolites, to indicate their presence, identity, or activity. In engineered detection systems, samples are processed to capture a target biomarker, then converted into a measurable signal through processes such as nucleic acid amplification, antibody binding, or bioreceptor-based sensing; signal strength can support qualitative or quantitative analysis. These methods support rapid diagnostics, environmental monitoring, food safety, and bioprocess control by improving pathogen screening and enabling earlier responses. Engineering advances focus on sensitivity, selectivity, portability, and integration into automated platforms.

2025

This video demonstrates a colorimetric E. coli detection method, involving an E. coli biomarker activating a DNAzyme, leading to cleavage of RNA linkage and detachment of a DNAzyme-urease conjugate. A subsequent colorimetric assay detects the E. coli biomarker through ammonia-induced pH changes.

A Capillary Electrophoresis Immunoassay for Protein Biomarker Detection

This video demonstrates a capillary electrophoresis immunoassay protocol for detecting a protein biomarker associated with amyotrophic lateral sclerosis (ALS) from human blood platelets. Confirmation of the presence of the ALS biomarker protein is achieved through size-based separation of the platelet proteins followed by chemiluminescent detection.

We describe an electrochemical sensor assay method for rapid bacterial detection and identification. The assay involves a sensor array functionalized with DNA oligonucleotide capture probes for ribosomal RNA (rRNA) species-specific sequences. Sandwich hybridization of target rRNA with the capture probe and a horseradish peroxidase-linked DNA oligonucleotide detector probe produces a measurable amperometric current.

Source: Barber, A. E., et al. Preparation and Application of a New Bacterial Biosensor for the Presumptive Detection of Gunshot Residue. J. Vis. Exp. (2019).This video demonstrates the detection of specific analytes using a bacterial biosensor. A bacterial culture carrying a plasmid with a red fluorescent protein (RFP) gene under an analyte-sensitive promoter is taken. After thoroughly wiping the test surface, a small section of the wipe is immersed in the culture and incubated. The analyte...
