Fully Transposed Line

A fully transposed line is a three-phase transmission line in which each conductor occupies every physical position along the route for equal lengths, an arrangement that balances the electrical properties of the phases. During transposition, conductors are shifted between positions at designated points, averaging differences in magnetic coupling, inductance, and capacitance caused by their spacing from one another and from ground. This balancing reduces voltage and current asymmetry, limits unwanted interference, and improves the accuracy of transmission-line models. Fully transposed lines are therefore important in power-system engineering, where stable three-phase operation and reliable analysis of long overhead transmission circuits are required.

Gene Trapping Using Gal4 in Zebrafish

This protocol describes the method of gene trap insertional mutagenesis using Gal4-VP16 as the primary reporter and GFP/RFP as secondary reporters in zebrafish. Approximately one in ten high-expressing F0 fish yield gene trap progeny co-expressing GFP and RFP. The screening procedure can be readily scaled to adapt to the size of the laboratory performing the insertional mutagenesis screen.

T cells expressing a CD19-specific chimeric antigen receptor (CAR) are infused as investigational treatment of B-cell malignancies in our first-in-human gene therapy trials. We describe genetic modification of T cells using the Sleeping Beauty (SB) system to introduce CD19-specific CAR and selective propagation on designer CD19+ artificial antigen presenting cells.

piggyBac Transposon System Modification of Primary Human T Cells

We describe a method to genetically modify primary human T cells with a transgene using the non-viral piggyBac transposon system. T cells modified to using the piggyBac transposon system exhibit stable transgene expression.
