Host Pupae Transplantation

Host pupae transplantation is an experimental technique that transfers a developing pupa or selected pupal tissue into a different host, allowing researchers to separate donor genetic effects from the host environment. During transplantation, the donor material is positioned within or alongside the host pupa and maintained as development proceeds, so interactions between transplanted tissue, host physiology, and developmental signals can be observed. In genetics, this approach helps investigate tissue autonomy, developmental regulation, cellular interactions, and the expression of inherited traits. It can also support studies of organ formation, reproductive development, and how genetic programs respond to a new physiological context.

However, the ability to target genes using RNAi during pre-adult stages is limited in the major human malaria vector Anopheles gambiae. We describe an RNAi protocol to reduce gene function via direct injection during pupal development.
