Car-nk Cells

CAR-NK cells are natural killer (NK) cells genetically engineered to express chimeric antigen receptors (CARs), combining innate immune activity with targeted recognition of diseased cells. The CAR binds a chosen surface antigen and activates signaling domains that trigger NK-cell degranulation, cytokine release, and perforin- and granzyme-mediated cytotoxicity, often without requiring conventional major histocompatibility complex presentation. In immunology and infection research, CAR-NK cells provide a platform for developing targeted cellular therapies against cancer and selected virus-infected cells. Their potential for precise recognition, scalable manufacturing, and reduced graft-versus-host reactivity supports continued investigation of safer, adaptable immunotherapies.

Here, we present a method to expand peripheral blood natural killer (PBNK), NK cells from liver tissues, and chimeric antigen receptor (CAR)-NK cells derived from peripheral blood mononuclear cells (PBMCs) or cord blood (CB). This protocol demonstrates the expansion of NK and CAR-NK cells using 221-mIL-21 feeder cells in addition to the optimized purity of expanded NK cells.
