Fluorogenic Substrate

A fluorogenic substrate is a nonfluorescent compound that becomes fluorescent after an enzyme modifies or cleaves it, providing a measurable signal of enzymatic activity. In the presence of the target enzyme, substrate conversion releases or activates a fluorescent product, whose intensity can be measured using fluorescence microscopy, plate readers, or flow cytometry. In immunology and infection research, fluorogenic substrates help detect pathogen-associated enzymes, monitor immune-cell activity, and quantify biochemical reactions in cells or clinical samples. Their sensitivity and compatibility with real-time measurements support applications such as diagnostic assay development, enzyme characterization, and evaluation of antimicrobial or immunomodulatory effects.

This video demonstrates an assay to screen for the proteolytic activity of proteases using fluorogenic peptides. The protease recognizes its cleavage site on the peptide, cleaving it and separating the quencher from the fluorophore, enabling its fluorescence emission. The fluorescence signal is detected and analyzed to check for the cleavage efficiency of different peptide variants.
