Interleukin 6 Expression

Interleukin 6 expression is the regulated production of the cytokine IL-6 by immune and nonimmune cells, an important process in inflammation, host defense, and tissue responses. Infection, tissue damage, and immune signaling can activate transcription of the IL6 gene, leading to IL-6 secretion and signaling through membrane-bound or soluble IL-6 receptors that engage gp130 and the JAK/STAT3 pathway. In immunology and infection research, measuring IL-6 expression helps characterize inflammatory responses, identify disease-associated biomarkers, and evaluate how pathogens or treatments alter cytokine networks. These analyses also support investigation of excessive IL-6 signaling in conditions such as cytokine-driven systemic inflammation.

To model how inflammatory signals influence neurodevelopment in genetically susceptible organisms, we present a protocol for microinjecting IL-6 into the duct of Cuvier (common cardinal vein) of zebrafish larvae, enabling controlled induction of inflammation and assessment of its developmental impact in the context of tuberous sclerosis complex (TSC).

This procedure describes a quick and easy workflow to introduce siRNA into difficult to transfect cell lines and follow gene expression by real-time PCR. Use of an automated cell counter, multi-well electroporation plate, and automated electrophoresis station provide quick and reliable results without the need for expensive robotic handling.
