Peripheral Blood Cd4

Peripheral blood CD4 refers primarily to CD4-positive T lymphocytes circulating in the blood, a key immune-cell population that coordinates adaptive immune responses and provides insight into immune status. These helper T cells recognize antigen-derived peptides presented by major histocompatibility complex class II molecules on antigen-presenting cells, then activate and direct other leukocytes through cell signaling and cytokine production. Measuring CD4 cell abundance and phenotype, often by flow cytometry, supports studies of host defense, immune dysfunction, and infection. In clinical and research settings, peripheral blood CD4 analysis is especially important for evaluating immune suppression and monitoring diseases such as HIV infection.

Selective Isolation of CD4+ Vδ1+ γδ T Cells from Human Peripheral Blood Mononuclear Cells

This video demonstrates a protocol for the magnetic labeling-based isolation of a low-concentration population of CD4+Vδ1+γδ T cells from human peripheral blood mononuclear cells (PBMCs). Using a combination of FITC-conjugated anti-Vδ1 antibodies, magnetic anti-FITC microbeads, and anti-CD4 antibody-coated superparamagnetic beads, the CD4+Vδ1+ T-cells can be successfully purified from the PBMC population.

Here, we present a protocol for the isolation of large plasma membrane-derived microvesicles from peripheral blood samples and their subsequent analysis by conventional flow cytometry and Western Blotting.

This video demonstrates a flow cytometry-based technique using two fluorochromes to identify CD4+ and CD8+ T cells, γδ T cells, B cells, natural killer (NK) cells, and monocytes in human peripheral blood mononuclear cells (PBMCs). Upon incubating the cells with fluorochrome-conjugated antibodies against the surface markers, the cell subsets are identified using an optimal flow cytometric gating strategy.

Generation and Culture of Blood Outgrowth Endothelial Cells from Human Peripheral Blood

This protocol allows for the reliable generation and characterization of blood outgrowth endothelial cells (BOECs) from a small volume of adult peripheral blood. BOECs can be used as a surrogate for endothelial cells from patients with vascular disorders and as a substrate for the generation of induced pluripotent stem cells.
