Rabbit Complement

Rabbit complement is the plasma protein system that provides rapid, innate immune defense and helps coordinate antibody-mediated responses during infection. Activated through classical, lectin, or alternative pathways, complement enzymes cleave C3 to generate fragments that promote opsonization and inflammation, while terminal components assemble the membrane attack complex on susceptible microbial membranes. In immunology and infection research, rabbit complement supports studies of pathogen susceptibility, immune complex clearance, and interactions between innate and adaptive immunity. Measuring or adding rabbit complement to experimental systems can clarify serum bactericidal activity and complement-dependent antibody effects, informing comparative immunology and assay design.

Complement System

2024

The complement system is a group of approximately 20 plasma proteins that strengthen the body's defenses against infections through opsonization, inflammation, and cell lysis. Opsonization involves coating pathogens with complement proteins, making them more recognizable and facilitating phagocyte engulfment. Certain complement proteins induce inflammation that attracts immune cells to the site of infection. Cell lysis involves the destruction of pathogens through the formation of a membrane...

Complementation Tests

A complementation test is a simple cross to identify whether the two mutations are located on the same gene or different genes. It was first performed by Edward Lewis in the 1940s while working on fruit flies. He developed the test to identify the location and arrangement of different mutations on chromosomes. Organisms heterozygous for different mutations are crossed pairwise in all combinations. If present on different genes, the mutations can complement each other by providing the missing...

Modeling Complement-Mediated Phagocytosis Using Opsonized Red Blood Cells In Vitro

Source: Nagre, N., et al. Alveolar Macrophage Phagocytosis and Bacteria Clearance in Mice. J. Vis. Exp. (2019).This video demonstrates the assessment of complement receptor-mediated phagocytosis using sheep red blood cells opsonized with IgM and complement, followed by their uptake by mouse macrophages. The extent of phagocytosis is quantified by measuring a blue chromogenic product formed through hemoglobin-catalyzed oxidation of diaminofluorene.

All animal procedures described here must be conducted in accordance with institutional animal ethics guidelines and approved by IACUC. All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Blood sampling is commonly performed in laboratory rabbits for research and clinical applications. The collection site is selected based on the required sample volume, sampling frequency, and study objectives. As a general...
