Cardiac Synchronicity Assessment

Cardiac synchronicity assessment is the evaluation of how precisely the heart’s chambers and myocardial regions activate and contract together, providing insight into cardiac performance. It combines electrical measurements, such as electrocardiography, with mechanical assessments from echocardiography, tissue Doppler, or strain imaging to identify delays between ventricular segments or between electrical activation and contraction. In medicine, these measurements help characterize dyssynchrony in heart failure, support evaluation of patients considered for cardiac resynchronization therapy, and monitor treatment-related changes. Standardized assessment also strengthens research on cardiac mechanics, disease progression, and therapies designed to improve coordinated pumping.

We describe here, the establishment and application of an Tg(Myh6-cre)1Jmk/J /Gt(ROSA)26Sortm38(CAG-GCaMP3)Hze/J (referred to as αMHC-Cre/Rosa26A-Flox-Stop-Flox-GCaMP3 below) mouse reporter line for cardiac reprogramming assessment. Neonatal cardiac fibroblasts (NCFs) isolated from the mouse strain are converted into induced cardiomyocytes (iCMs), allowing for convenient and efficient evaluation of reprogramming efficiency and functional maturation of iCMs via calcium (Ca2+) flux.

Assessment of Cardiac Function and Energetics in Isolated Mouse Hearts Using 31P NMR Spectroscopy

Langendorff-mode isolated heart perfusion, in conjunction with 31P NMR spectroscopy, combines the fields of biochemistry and physiology into one experiment. The protocol allows for the dynamic measurement of high energy phosphate content and turnover in the heart while concurrently monitoring physiologic function. When performed correctly, this is a valuable technique in the assessment of cardiac energetics.
