Cold Ischemia Time

Cold ischemia time is the interval during which an organ or tissue remains without blood flow while preserved at a low temperature before transplantation or clinical use. Cooling, typically with specialized preservation solutions, suppresses cellular metabolism and reduces oxygen demand, but does not stop hypoxia-related injury, ATP depletion, and membrane damage. In transplant medicine, limiting cold ischemia time helps preserve graft viability and supports early organ function after reperfusion. Measuring and managing this interval guides organ allocation, transport, preservation strategies, and surgical planning, while also informing research on machine perfusion and other approaches designed to extend safe preservation.

A protocol for stroke onset time estimation in a rat model of stroke exploiting quantitative magnetic resonance imaging (qMRI) parameters is described. The procedure exploits diffusion MRI for delineation of the acute stroke lesion and quantitative T1 and T2 (qT1 and qT2) relaxation times for timing of stroke.

Source: Robert M. Rioux & Suprita Jharimune, Pennsylvania State University, University Park, PA Working with extreme temperatures, both high and low, is an integral part of many laboratory operations. For many, mentioning a laboratory instantly evokes the mental picture of a Bunsen burner. Bunsen burners and hot plates are used extensively in small and large operations in research laboratories and industries, thus making it necessary for all users to be aware of their safe handling...

Digital intravital epifluorescence microscopy of postcapillary venules in the cremasteric microcirculation is a convenient method to gain insights into leukocyte-endothelial interaction in vivo in ischemia-reperfusion injury (IRI) of striated muscle tissue. We here provide a detailed protocol to safely perform the technique and discuss its applications and limitations.
