Full Thickness Excision

Full thickness excision is a surgical procedure that removes a lesion or area of abnormal tissue through the entire thickness of the skin and often into the underlying subcutaneous tissue. The surgeon makes an incision around the target, separates it from surrounding structures, and removes an intact specimen that includes the epidermis, dermis, and deeper tissue; the wound may then be closed directly or repaired with a graft or flap. In medicine, this approach supports complete lesion removal while preserving tissue for histopathological examination. It is used for selected skin tumors, suspicious lesions, and other conditions requiring assessment of tissue depth, margins, and disease extent.

Here we describe a cost-effective technique for organotypic culture of adult porcine retina for seven days. Briefly, a sterile filter paper was used to lift the neural retina off from the RPE and place photoreceptor side up on an insert raised by a custom-made stand.
