Ischemia Time Measurement

Ischemia time measurement is the determination of how long tissue or an organ remains deprived of adequate blood flow and oxygen, a time interval that strongly influences clinical assessment and treatment. Clinicians establish timestamps for the onset of interrupted perfusion and for revascularization or reperfusion, distinguishing warm ischemia at body temperature from cold ischemia during preservation or storage. In medicine, these measurements support decisions in emergency care, surgery, and organ transplantation, where prolonged ischemia can worsen cellular injury and impair graft function. Accurate timing also standardizes communication, enables comparison of outcomes, and helps researchers evaluate strategies for limiting ischemic damage.

A Magnetic Resonance Imaging Protocol for Stroke Onset Time Estimation in Permanent Cerebral Ischemia

A protocol for stroke onset time estimation in a rat model of stroke exploiting quantitative magnetic resonance imaging (qMRI) parameters is described. The procedure exploits diffusion MRI for delineation of the acute stroke lesion and quantitative T1 and T2 (qT1 and qT2) relaxation times for timing of stroke.

Digital intravital epifluorescence microscopy of postcapillary venules in the cremasteric microcirculation is a convenient method to gain insights into leukocyte-endothelial interaction in vivo in ischemia-reperfusion injury (IRI) of striated muscle tissue. We here provide a detailed protocol to safely perform the technique and discuss its applications and limitations.
