Microvascular Density

Microvascular density is a measure of the number or spatial distribution of small blood vessels within a defined tissue area, providing insight into local blood supply and vascular organization. It is assessed by identifying microvessels in tissue sections or imaging data, often using endothelial markers, and calculating vessel counts or vessel area relative to the sampled tissue. In medicine, microvascular density helps characterize angiogenesis, tumor biology, wound healing, and ischemic disease. Changes in this measure can indicate altered perfusion, support disease classification and prognosis, and inform research into therapies that promote or inhibit blood-vessel formation.

Here, we describe a methodology to deliver human cord blood-derived endothelial colony-forming cells (ECFCs) and bone marrow-derived mesenchymal stem cells (MSCs), embedded in a collagen/fibronectin gel, subcutaneously into immunodeficient mice. This cell/gel combination generates a human vascular network that connects with the mouse vasculature.

There are many available options for management of the patient with trigeminal neuralgia. Microvascular decompression, while the most invasive of all options, is also the most effective at achieving long term remission of symptoms. Video instruction on how to maximize efficacy and minimize complications with this procedure is described.

Density Gradient Ultracentrifugation

2023

Density gradient ultracentrifugation is a common technique used to isolate and purify biomolecules and cell structures. This technique exploits the fact that, in suspension, particles that are more dense than the solvent will sediment, while those that are less dense will float. A high-speed ultracentrifuge is used to accelerate this process in order to separate biomolecules within a density gradient, which can be established by layering liquids of decreasing density in a centrifuge tube. The...

Generating an Ultra-Low-Density Neuronal Culture Using a High-Density Neuronal Feeder Layer

2025

This video demonstrates the method for culturing ultra-low-density neurons in the presence of a high-density neuronal feeder layer. It establishes a co-culture of varying-density neurons, ensuring close physical proximity. The growth factors secreted by high-density neurons help neuronal survival and growth, maintaining ultra-low-density neurons for a longer time period.
