Wharton Duct Cannulation

Wharton Duct Cannulation is a minimally invasive technique for accessing the submandibular (Wharton) duct, the channel that carries saliva from the submandibular gland into the mouth. It typically involves locating and gently dilating the ductal papilla beside the lingual frenulum before advancing a fine cannula into the duct under direct or endoscopic guidance. This access can permit irrigation, contrast injection, stone retrieval, or passage of a sialendoscope to evaluate strictures and other causes of obstructed salivary flow. In medicine, the technique supports minimally invasive diagnosis and treatment of obstructive sialadenitis, helping preserve gland function and reduce the need for open surgery.

A protocol for the cannulation of the mouse submandibular salivary gland via the Wharton's duct is described. For this experiment, the trypan blue solution is used as a dyer to demonstrate how this technique effectively delivers infusions into the targeted gland, and to suggest the reliability of this new approach as a potential clinical drug/cell therapy for the regeneration of salivary glands.

Here we describe a technique to cannulate the mesenteric lymph duct in rats which enables quantification of lipid and drug transport via the lymphatic system following intestinal delivery. The technique can be adapted to assess mesenteric lymph concentrations and/or transport of fluid, immune cells, peptides, proteins and lipophilic molecules.

This video demonstrates intraduodenal cannulation in a rat model. After anesthetizing the rat, the duodenum is located and a small puncture is created. A J-shaped cannula with a beveled tip is carefully inserted and secured, and the incision is closed. The cannulated rat model enables intra-intestinal infusion for immunopathology studies.
