Enforced Locomotion Treadmill

An enforced locomotion treadmill is a motorized apparatus that compels an animal to walk or run at a controlled pace, enabling precise study of movement and neural control. As the belt moves beneath the subject, it must generate continuous stepping to maintain its position, while researchers adjust speed and other conditions to assess locomotor responses. In neuroscience, this method supports investigation of motor circuits, sensory feedback, gait coordination, and locomotor adaptation. It also provides a reproducible platform for evaluating changes caused by neural injury, genetic manipulation, disease, or experimental treatments, helping link behavior with underlying nervous-system function.

We describe a protocol for investigating human locomotor adaptation using the split-belt treadmill, which has two belts that can drive each leg at a different speed. We specifically focus on a paradigm designed to test the generalization of adapted locomotor patterns to different walking contexts (e.g., gait speeds, walking environments).

The Treadmill Fatigue Test: A Simple, High-throughput Assay of Fatigue-like Behavior for the Mouse

Fatigue is a common, undertreated and frequently poorly-understood symptom in many diseases and disorders. New preclinical assays of fatigue may help to improve current understanding and future treatment of fatigue. To that end, the current protocol provides a novel means of measuring fatigue-like behavior in the mouse.
