Enhancer Testing

Enhancer testing is the experimental evaluation of DNA sequences that can increase transcription from a gene, helping researchers determine how regulatory elements control cell-specific gene expression. In a typical assay, candidate enhancer sequences are placed near a reporter gene and introduced into cells; reporter output is then compared across conditions to determine whether the sequence activates transcription, often during neuronal differentiation or in response to signaling. In neuroscience, these tests help map regulatory programs underlying neuronal identity, brain development, and disease-associated variants, while supporting functional interpretation of noncoding genomic regions.
