Dura Mater Injection

Dura mater injection is a neuroscience technique that delivers a solution into or immediately adjacent to the dura mater, the tough outer layer of the meninges surrounding the brain and spinal cord. By placing an experimental compound at this meningeal interface, researchers can examine how local chemical, inflammatory, or pharmacological signals affect nearby sensory nerve fibers and nervous-system activity. The method supports studies of meningeal biology, headache mechanisms, neuroinflammation, and pain signaling, while providing a controlled way to evaluate candidate treatments or trace responses in tissues that are difficult to access through conventional systemic administration.

Here we present a protocol to visualize spatial correlation of calcitonin gene-related peptide (CGRP)-immunoreactive nerve fibers and blood vessels in the cranial dura mater using immunofluorescence and fluorescent histochemistry with CGRP and phalloidin, respectively. In addition, the origin of these nerve fibers was retrograde traced with a fluorescent neural tracer.
