Flim Acquisition

FLIM acquisition is the process of collecting fluorescence lifetime measurements from labeled molecules or structures, revealing how long fluorophores remain excited rather than simply how bright they appear. Typically, a pulsed light source excites fluorescent probes, and detectors record the arrival times of emitted photons to calculate lifetimes through time-correlated single-photon counting or related timing methods. In neuroscience, these measurements can map molecular environments, monitor ion-sensitive or voltage-sensitive indicators, and distinguish biochemical interactions within neurons and neural circuits. Because fluorescence lifetime is less dependent on probe concentration and excitation intensity than fluorescence intensity, FLIM acquisition supports quantitative imaging in complex living tissue.
