Fluorescent Reporter Protein

A fluorescent reporter protein is a genetically encoded molecule that produces visible fluorescence, allowing researchers to track gene expression, cell identity, or protein localization in living systems. Typically derived from proteins such as green fluorescent protein, it forms an internal chromophore that emits light at a characteristic wavelength after excitation, and its gene can be linked to a promoter or target protein. In neuroscience, fluorescent reporters help reveal neuronal morphology, monitor circuit development, and identify cells activated under specific conditions. Their compatibility with microscopy enables spatial and, in some cases, temporal analysis of neural activity, supporting studies of brain organization, connectivity, and disease mechanisms.

The present protocol describes a procedure to perform fluorescent size exclusion chromatography (FSEC) on membrane proteins to assess their quality for downstream functional and structural analysis. Representative FSEC results collected for several G-protein coupled receptors (GPCRs) under detergent-solubilized and detergent-free conditions are presented.
