Brain Hemisphere Securing

Brain hemisphere securing is the controlled stabilization of one or both cerebral hemispheres during anatomical preparation, imaging, or experimental manipulation. The procedure uses physical support or fixation to limit movement, preserve the brain’s orientation, and reduce deformation while researchers examine neural structures and pathways. In neuroscience, reliable securing improves the consistency of dissections, sectioning, microscopy, and other laboratory procedures that require precise access to cortical and subcortical regions. By maintaining tissue position throughout an experiment, it supports clearer anatomical interpretation, more reproducible measurements, and safer handling of delicate brain specimens.

This video demonstrates in vivo recording of local field potential (LFP) from bilateral M2 areas of the mouse brain. An anesthetized mouse is secured and prepared for the procedure. To record LFPs, insert microelectrodes into the target coordinates of the bilateral M2 areas, which are responsible for complex motor movements.

We applied repetitive transcranial magnetic stimulation (rTMS) to the unilateral hemisphere of rat brain, by placing a 25-mm figure-8 coil 1 cm lateral to the vertex on the biauricular line and angulating the coil by 45°. An in-house water cooling system was used for rTMS for more than 20 min.
