Glass Capillary Method

The Glass Capillary Method is a microscale laboratory technique that uses a finely drawn glass tube to access, measure, or manipulate individual cells, making it valuable for investigating neuronal function. A glass capillary is heated and pulled into a narrow tip, then filled with conductive solution or connected to a pressure system; its tip can penetrate a cell, form a high-resistance seal with the membrane, record electrical signals, or deliver small amounts of material. In neuroscience, the method supports intracellular recording, patch-clamp experiments, microinjection, and localized stimulation. These applications reveal membrane properties, synaptic activity, and cellular responses with high spatial precision.

Capillaroscopy is a non-invasive, efficient, relatively inexpensive and easy-to-learn methodology for directly visualizing capillaries in the microcirculation. However, only one publication to date describes the reliability of a complex software program available for quantitating capillaroscopy data. Here, we present a simple, reliable protocol for quantitating capillaries using a standardized algorithm.

2023

This video describes the CApillary FEeder, or CAFE, assay, a behavioral method that measures food intake and preference of unrestrained fruit flies. The featured protocol shows how to perform the assay with minimal evaporation from the liquid-filled capillaries used to deliver the food and track its intake.
