Image Subtraction

Image subtraction is a computational technique that compares two images by removing corresponding pixel values, making changes between them easier to detect. In neuroscience, researchers typically register a baseline and experimental image, then subtract the reference frame pixel by pixel to reduce shared background features and produce a difference image. This process can highlight changes in fluorescence, cellular morphology, or tissue structure that may be difficult to see in raw data. Image subtraction supports analysis of neural activity, microscopy experiments, and other time-resolved imaging studies by separating stimulus-related or disease-associated changes from relatively stable visual information.

Rapid Subtractive Patterning of Live Cell Layers with a Microfluidic Probe

We present a protocol to perform subtractive patterning of live cell monolayers on a surface. This is achieved by local and selective lysis of adherent cells using a microfluidic probe (MFP). SOM underlies many soil functions and processes, but its characterization by FTIR spectroscopy is often challenged by mineral interferences. The described method can increase the utility of SOM analysis by FTIR spectroscopy by subtracting mineral interferences in soil spectra using empirically obtained mineral reference spectra.
