Nerve Reinnervation

Nerve reinnervation is the restoration of neural connections between regenerating nerves and their target tissues after injury, supporting the recovery of sensation or movement. In the peripheral nervous system, surviving axons can extend through guided pathways, with Schwann cells and associated signaling molecules promoting axonal growth toward denervated muscles, glands, or sensory receptors; functional recovery depends on accurate reconnection and synapse formation. Neuroscience research examines reinnervation to understand neural repair and improve outcomes after trauma, surgery, or disease. These principles inform nerve grafting, reconstructive procedures, rehabilitation strategies, and emerging bioengineered or neuroprosthetic approaches.

Source: Svientek, S. R., et al. Fabrication of the Composite Regenerative Peripheral Nerve Interface (C-RPNI) in the Adult Rat. J. Vis. Exp. (2020).This video demonstrates the surgical construction of a composite regenerative peripheral nerve interface (C-RPNI) in a rat model of peripheral nerve injury. The procedure involves attaching a transected peroneal nerve to a muscle graft and overlaying it with a dermal skin graft to promote nerve regeneration. Over time, motor and sensory neurons...

In this report we describe a method to crush mouse sciatic nerve. This method uses readily available hemostatic forceps and easily and reproducibly produces complete sciatic nerve crush. In addition, we describe a method to prepare muscle whole mounts suitable for analysis of nerve regeneration after sciatic nerve crush.
