Neuroinflammatory Changes

Neuroinflammatory changes are alterations in the nervous system caused by immune activity within or around neural tissue, often influencing neuronal function and disease progression. They arise when microglia and astrocytes detect injury, infection, or cellular stress and release signaling molecules such as cytokines, while changes in the blood-brain barrier can modify immune-cell access to the brain. This response may support tissue repair when controlled but can disrupt synaptic signaling and damage neurons when persistent. Studying neuroinflammatory changes helps researchers understand conditions including multiple sclerosis, Alzheimer’s disease, Parkinson’s disease, and traumatic brain injury, while guiding biomarker discovery and development of targeted therapies.

Levels of the inflammatory cell-signaling molecule nitric oxide (NO) are commonly assayed using Griess reagent. In this protocol, we have created a modified Griess assay utilizing live Drosophila brain tissue in order to detect the secretion of NO in a simple, quantifiable and highly repeatable method.

Mouse Footpad Inoculation Model to Study Viral-Induced Neuroinflammatory Responses

The footpad inoculation model is a valuable tool for characterizing viral-induced neuroinflammatory responses in vivo. In particular, it provides a clear assessment of viral kinetics and associated immunopathological processes initiated in the peripheral nervous system.

Detection of Cerebral Microhemorrhages in a Rat Model

2025

This video demonstrates an assay to detect Cerebral microhemorrhages (CMHs) in a rat model. CMHs are brain bleeds due to blood-brain barrier disruption, identified by leaked red blood cells (RBCs). In a rat model, fixed brain tissue is cryoprotected, sectioned, and stained with hematoxylin-eosin for RBCs, allowing microscopic visualization of CMHs. Leaked RBCs appear as red-orange under the microscope.

We present a protocol to investigate the mRNA expression biomarkers of periosteum-derived cells (PDCs) induced by vitamin C (vitamin C) and 1,25-dihydroxy vitamin D [1,25-(OH)2D3]. In addition, we evaluate the ability of PDCs to differentiate into osteocytes, chondrocytes, and adipocytes.
