Primary Astrocytes

Primary astrocytes are non-immortalized glial cells isolated directly from central nervous system tissue and maintained in culture to study astrocyte biology in a physiologically relevant context. They help regulate the neural environment by buffering extracellular ions, removing neurotransmitters such as glutamate, providing metabolic support, and altering gene expression and morphology in response to injury or inflammatory signals. In neuroscience research, primary astrocyte cultures are used to investigate neuron-glia communication, neuroinflammation, neurotoxicity, and disease-associated cellular responses. Because they retain many properties of tissue-derived cells, these cultures can complement immortalized lines and support mechanistic studies and preclinical evaluation of therapeutic strategies.

Generating a Primary Culture of Astrocytes from a Rat Pup Brain

The video demonstrates a technique for culturing primary astrocytes from the rat cortex. The cortex is mechanically dissociated to obtain a homogeneous glial cell suspension, which is then plated on polypeptide-coated plates to promote selective proliferation of astrocytes. Subsequently, the astrocytes are enzymatically dissociated from the plates and transferred to fresh media, allowing the culture of pure astrocytes.
