Protein Isolation Method

Protein isolation methods are laboratory techniques used to separate target proteins from cells, tissues, or complex biological mixtures for biochemical and functional analysis. In neuroscience, the process typically combines tissue homogenization and cell lysis with centrifugation, selective solubilization, or chromatography, using differences in solubility, size, charge, or affinity to enrich the protein of interest while limiting degradation through controlled conditions and protease inhibition. Isolated neuronal proteins can then be examined by electrophoresis, immunodetection, mass spectrometry, or activity assays to investigate signaling pathways, synaptic function, disease-associated changes, and protein structure. Reliable isolation supports reproducible research into brain biology and therapeutic targets.

A tool and chemistries are described to sequentially isolate nucleic acids followed by proteins from a sample without the need for electricity. The tool consists of a sorbent held within a transfer pipette while the isolation chemistries are based on solid-phase extraction principles.
