Vasodilation Mechanisms

Vasodilation mechanisms are the physiological and pharmacological processes that widen blood vessels, lowering vascular resistance and altering tissue blood flow. They act primarily by relaxing vascular smooth muscle through pathways such as nitric oxide activation of guanylyl cyclase and cyclic GMP production, increased cyclic AMP signaling, reduced intracellular calcium, or inhibition of contractile receptors. In pharmacology, drugs that target these pathways include nitrates, calcium channel blockers, potassium channel activators, and alpha-adrenergic antagonists. Understanding these mechanisms supports the treatment of hypertension, angina, heart failure, and circulatory disorders while helping researchers predict effects on blood pressure, perfusion, and adverse responses.

We describe the isolation of cardiac extracellular matrix from C57Bl/6J control mice, tight-skin mice, and tight-skin mice treated with the IRF5 inhibitory peptide. We also describe the vasodilation studies on the isolated vessels from C57Bl/6J, tight-skin mice and tight-skin mice treated with the IRF5 inhibitory peptide.

Antihypertensive Drugs: Vasodilators

2024

Vasodilators, primarily affecting the smooth muscles within arterial and venous walls, are commonly used for hypertension treatment. Medications such as minoxidil and hydralazine primarily target arteries and arterioles, while sodium nitroprusside acts on arterioles and venules. Minoxidil, functioning as a prodrug, is metabolized by hepatic sulfotransferase into its active form, minoxidil sulfate, after oral administration. This metabolite binds to the sulfonylurea receptor (SUR) component of...

The biosynthesis of cartilaginous extracellular matrix by chondrocytes can be affected by application of mechanical stimuli. This method describes the technique of applying dynamic compressive strains to chondrocytes encapsulated in 3D constructs and the evaluation of induced changes in chondrocyte metabolism.
