전남대학교병원 실험동물연구센터 윤리위원회(승인번호. CNU IACUC - H - 2022-36)은 모든 동물 실험을 승인했습니다. 본 연구에 사용된 수컷 Sprague-Dawley 랫트(350-450 g)는 실험동물의 관리 및 사용에 대한 가이드라인에 따라 관리를 받았다. 쥐는 12시간 명암 주기로 온도 조절이 가능한 방에 수용되었으며 표준 음식과 물을 사용할 수 있었습니다.
1. 준비
참고: 한 명의 실험자가 모든 실험 절차를 수행할 수 있습니다.

</ol>2. 기증자 심장 보존 및 채혈
<ol><li>마취실에서 이소플루란(5%)으로 기증자 쥐의 마취를 유도하고 쥐의 체중을 기록한 후 수술대에 올려 놓습니다.</li>
	<li>쥐를 수술대 위의 앙와위 자세로 놓고 노즈콘을 통해 90% 산소와 함께 2%-2.5% 이소플루란을 전달하여 지속적인 마취를 시행합니다.</li>
	<li>발가락 핀치에 대한 반응 부족과 분당 50-60 사이 여야하는 호흡 빈도를 확인하여 마취 깊이를 확인하십시오. 참고: 적절한 수준의 마취는 기증자 쥐에게 불필요한 스트레스와 통증을 피하는 데 중요합니다.</li>
	<li>눈 윤활제를 바르고 수술이 수행될 쇄골에 치골 부위를 면도합니다. 요오드 기반 스크럽과 70% 알코올로 해당 부위를 청소하십시오.</li>

</ol>3. 현장 관류
<ol><li>대동맥 캐뉼라 삽입 및 관류<ol><li>대동맥 캐뉼러를 삽입하기 전에 식염수 프라이밍 회로를 혈액 프라이밍으로 교체하십시오.</li>

		<li>저장소에서 0.15mL의 혈액을 수집하고 관류 시작 시와 그 후 20분마다 혈액 가스 분석(BGA)을 확인합니다. 관류 동안 pH, pCO2,pO2, 글루코스, 헤마토크릿, 칼륨 및 젖산염을 모니터링하고 기록합니다. 120분 관류 후 주사기 펌프를 통해 3mL의 Custodiol을 250mL/h의 속도로 투여하여 심장을 정지시킵니다.</li>

</ol>4. 이식
<ol><li>수혜자의 준비<ol><li>현장 관류가 중단되기 30분 전에 수용자 준비를 시작합니다.</li>
		<li>2.2 단계에서 언급 한 것과 동일한 방법을 사용하여 수용자 동물을 마취시킵니다.</li>
		<li>쥐를 가열 패드에 앙와위 자세로 놓고 온도 프로브를 직장에 삽입하여 체온을 37°C로 유지합니다.</li>

	</ol></li>
	<li>중간 선 개복술을 수행하고 복강을 넓히기 위해 견인기를 삽입하십시오. 면봉을 사용하여 수혜자의 왼쪽으로 복부 장기를 동원하여 시술을 위한 공간을 만듭니다.</li>
	<li>복부 장기를 따뜻하고 젖은 거즈로 감싸서 탈수를 예방하십시오. 수술 중 50mL 주사기로 따뜻한 식염수를 간헐적으로 뿌립니다.</li>
	<li>10배 배율의 수술용 현미경을 사용하여 십이지장과 근위 공장을 면봉으로 둔기로 해부하여 Abd. A. 및 IVC를 노출시킵니다. Abd를 준비합니다. A 및 IVC를 문합하고, 도 3 또는 이전에 문서화된 방법15에 따라 기증자 심장을 체계적으로 이식한다. 알림: Abd. A.와 IVC를 분리하지 마십시오.<ol><li>혈관 문합이 신장 내하에 배치된다고 가정하면 클램핑을 위해 대동맥과 IVC의 충분한 부분을 준비하십시오.</li>

Rat Model of Normothermic Ex-Situ Perfused Heterotopic Heart Transplantation
