Mouse Round Spermatid Injection

우리는 AOA가 배아의 발달 능력에 미치는 영향을 조사하는 것으로 조사를 시작했습니다. 실험 설계의 개략도는 그림 1A에 나와 있습니다. 정자 주사 전에 난모세포는 AOA(A-ICSI)를 거치거나 치료되지 않은 상태(ICSI)를 받았습니다. 배아 발달에 대한 자세한 데이터는 표 1에 제시되어 있습니다. 그 결과, A-ICSI군과 ICSI 군 간에 분열, 배반포 또는 출생률에 유의한 차이가 없는 것으로 나타났다(P &gt; 0.05; 그림 1B). 이러한 결과는 10mM SrCl2를 20분 동안 사용하는 AOA가 배아의 발달 잠재력에 영향을 미치지 않았음을 나타냅니다.
이전에 FACS 또는 현미경 하의 직접 육안 검사에 의해 선택된 ROSI 배아의 발달 효율에는 눈에 띄는 차이가 없다고 보고되었습니다22. 우리의 실험에서는 RS를 식별하기 위해 FACS 기술을 사용했습니다(그림 2A, B). 현미경으로 보니 생쥐의 둥근 정자는 직경이 약 10μm였으며 중간에 돌출부와 같은 핵소체 구조를 보여주었습니다(그림 2C). 우리는 FACS를 통한 선택이 현미경으로 직접 육안 검사하는 것보다 더 정확하다고 가정합니다. 또한, 문헌은 형태학적 차이를 통한 RS의 직접적인 탐색을 뒷받침합니다.
