Preparing Retinal Organoid Samples for Transmission Electron Microscopy

iPSC 분화를 통한 3D RO의 확립은 망막 질환 메커니즘 및 줄기세포 대체 요법을 연구하기 위한 강력한 도구를 제공합니다. 다른 연구자들이 RO의 시냅스 연결을 기능적 및 면역세포화학적으로 입증했지만, 기존 시냅스와 리본 시냅스의 직접적인 증거는 매우 제한적입니다. 여기에서는 TEM에 의해 RO에서 두 가지 유형의 시냅스의 미세 구조를 조사하는 방법을 제시합니다. 180일 배양 후 RO를 고정, 염색, 매립 및 초박형 슬라이스했습니다. TEM 관찰은 OPL에 분포된 광수용체(그림 1 및 그림 2)의 축삭 말단에서 리본 시냅스를 밝혀냈습니다. TEM에서는 간상체의 리본이 시냅스 침입 주위를 구형으로 둘러싸고 수평 세포가 있는 말굽 모양의 프로필을 형성합니다(그림 1). 막대에는 일반적으로 하나의 리본만 있지만 원뿔에는 여러 개의 리본이 있습니다(그림 2). 또한, IPL의 무축삭 세포 사이의 고전적인 화학적 시냅스가 관찰되었습니다(그림 3). TEM에서 화학적 시냅스는 시냅스전(presynaptic), 시냅스 틈새(synaptic cleft), 시냅스후(postsynaptic)의 세 부분으로 구성됩니다. 시냅스전(presynaptic) 부분과 시냅스후(postsynaptic) 부분에 해당하는 세포막은 나머지 부분보다 약간 두껍고, 두 막 사이의 좁은 틈을 시냅스 틈(synaptic cleft)이라고 합니다. 시냅스전막 부위의 세포질에는 많은 시냅스 소포가 있습니다. 이 연구는 iPSC에서 파생된 3D RO에서 완전한 시냅스 연결의 존재를 확인합니다. 그러나 단계 사이에 불완전한 헹굼은 시료 준비 과정에서 검은색 입자가 침전될 수 있다는 점에 유의해야 합니다(그림 4).
