Acute Single-Unit Multi-Electrode Recordings from the Brainstem of Head-Fixed Mice

5명의 남성 C57BL/6J에게 ECoG 헤드셋과 헤드플레이트를 이식했습니다(그림 4A). 회복 후, 마우스는 서로 다른 날에 두 번의 1.5시간 세션 동안 머리 고정과 전기생리학 기록 장비에 습관화되었습니다(그림 4B). 다음으로, 2mm x 2mm 개두술 창을 만들고(그림 4C) 마우스를 깨우고 머리를 고정한 상태에서 실리콘 프로브를 삽입했습니다(그림 4D). 두 가지 유형의 실리콘 UCLA 프로브가 사용되었습니다 : (1) 64 M : 1 섕크, 64 전극 프로브; (2) 256 ANS: 4 섕크, 256 전극 프로브. 세보플루란(Sevoflurane)과 산소는 전기생리학 장비에 고정된 노즈콘(nose cone)을 사용하여 투여하였다. 산소 및 세보플루란 수치는 인라인 가스 분석기를 사용하여 5분마다 모니터링했습니다. 경활 관류 후, 뇌를 채취하고(그림 4E) 프로브 궤적에 대해 평가했습니다(그림 4F). 그런 다음 기록된 활동 전위를 단일 단위 데이터로 선별했습니다(그림 4G).
MATLAB23에서 개발된 사용 가능한 프로브 재구성 알고리즘으로 프로브 배치를 검증하는 5개의 기록이 수행되었습니다. 이러한 알고리즘을 사용하여 실리콘 프로브의 각 전극을 특정 뇌 구조에 국한시켜 뇌 내 프로브 배치에 대한 3차원 관점을 획득했습니다(그림 5). 이러한 재구성 알고리즘은 프로브 궤적을 추가로 검증하기 위해 면역조직화학적 마커와 결합되었습니다(그림 6). 그런 다음 데이터를 선별된 단일 단위와 일치시켰습니다(그림 7).
총 64개의 단일 뉴런이 포착되었으며, 그 중 13개는 vlPAG에 위치했습니다. 나머지 51 개의 뉴런은 중뇌, 중뇌 망상핵, paratrochlear nucleus, cuneiform nucleus, laterodorsdal tegmental nucleus, parabrachial nucleus 및 superior cerebellar peduncles와 같은 인근 핵에있었습니다. 각 마우스에서 뉴런의 발화 활동은 히트 맵으로 표시됩니다(그림 7). 마우스 3은 단일 장치 기록 품질이 좋지 않아 분석에서 제외되었습니다. 대부분의 기록된 뉴런은 세보플루란 마취 중에 발화를 감소시켰다.
vlPAG 뉴런의 평균 발화는 5분 기준선(즉, 기록 10-15분) 및 세보플루란 중 5분(즉, 기록 40-45분)에 각 마우스에 대해 비교되었습니다. vlPAG 발화는 현저히 감소했으며(그림 8A), 더욱이 이러한 증가는 모든 vlPAG 뉴런에서 일관되게 나타났습니다(그림 8B).
