2. 오로보로스 일반 설정
<ol>
	<li>DatLab 소프트웨어를 시작합니다.</li>
	<li>실험 온도(37°C)와 교반 회전 속도를 챔버 sV에서 550rpm(9.2Hz 또는 초당 회전수) 또는 챔버 cV에서 750rpm(12.5Hz)으로 설정합니다. 데이터 샘플링 간격을 2초로 설정하여 기기 O2 백그라운드 테스트에서 80초(40개 데이터 포인트) 또는 호흡 측정의 경우 40초(20개 데이터 포인트)에 걸쳐 평활화합니다. 참고: 마그네틱 교반기 바(폴리비닐리덴 디플루오라이드[PVDF]로 코팅된 AlNiCo; 표 1) 챔버의 교반기 수준에서 3.5mT 전계 강도를 갖는 펄스 회전 자기장에 의해 교반됩니다.</li>
	<li>MiR05 또는 후속 실험에 사용된 것과 동일한 배양 배지에서 공기 보정28 을 수행합니다.</li>
	<li>실험을 시작하기 직전에 제로 산소 교정을 가끔 수행하지 마십시오.<ol>

3. 계측 O2 배경 테스트

4. Oroboros 챔버에 샘플 추가
참고: 샘플은 부분 부피 대체(본 연구에서 혈소판 및 섬유아세포) 또는 전체 부피 대체(본 연구에서 분리된 미토콘드리아)의 두 가지 방법으로 챔버에 추가할 수 있습니다.

	</li>

	</li>
	<li>살아있는 세포: 인간 혈소판<ol>
		<li>혈소판이 포함된 냉동 바이알을 37°C의 수조에 삽입하고 얼음이 거의 남지 않을 때까지 내용물을 해동합니다.</li>
		<li>냉동 바이알을 열고 예열된 배지 1mL(DPBS + 10mM EGTA, 37°C)를 천천히 방울로 추가합니다.</li>
		<li>혈소판을 부드럽게 혼합하고 총 부피 10mL의 예열 배지(DPBS + 10mM EGTA, 37°C)가 있는 50mL 원심분리기 튜브로 옮깁니다. 스윙 아웃 로터(가속 6, 브레이크 2)를 사용하여 실온(RT)에서 1000g 에서 10분 동안 원심분리합니다.</li>

	</li>
	<li>분리된 미토콘드리아: 마우스 심장<ol>
		<li>15μL의 미토콘드리아 현탁액을 5985μL의 미토콘드리아 호흡 배지(MiR05)에 첨가하면 0.013-0.026mg∙mL-1 범위의 스톡 미토콘드리아 단백질 농도가 됩니다.</li>
		<li>완전한 부피 교체를 통해 미토콘드리아 현탁액을 Oroboros 챔버에 추가합니다. 참고: 하나의 Oroboros는 각각 위치 A와 B에 sV 및 cV 챔버가 장착되었으며, 각각 위치 B와 A에 sV 및cV 챔버가 장착된 다른 Oroboros와 병렬로 사용되었습니다.</li>
