Optimized Intravenous Injection in Adult Zebrafish

성체 캐스퍼 피쉬에서 다양한 주입 방법의 효과와 정밀도를 평가하기 위해 심장 내, RO 및 IV 기술을 사용하여 비교 분석을 수행했습니다(그림 2A). Tg(mpo: EGFP) 어류의 다양한 WKM 세포를 주입하고, 각각을 1 × 105 및 3 × 105 세포의 농도로 FITC-덱스트란 염료와 혼합했습니다. 각 방법의 성공은 주입 후 GFP 신호 존재의 즉각적인 현미경 평가를 통해 결정되었습니다. 그 결과 여러 가지 방법으로 전달 성공률에 상당한 차이가 있는 것으로 나타났습니다. IV 주입 방법은 43마리의 물고기 중 41마리에서 GFP 신호가 감지되어 가장 높은 효과를 보여주었습니다. 대조적으로, RO 방법은 37마리의 물고기 중 3마리에서만 성공적인 주입이 관찰되는 등 제한적인 성공을 보였습니다. 심장 내 방법은 40마리의 물고기 중 11마리에서 성공적인 분만을 달성했습니다(그림 2A). 이식된 물고기의 생존 여부도 모니터링했습니다. RO 및 심장 내 방법을 통해 주입된 물고기는 정맥 주사된 물고기에 비해 생존율이 현저히 낮았습니다(그림 2B). 특히, IV를 주입한 물고기의 생존율은 IV를 주입한 세포의 수에 영향을 미쳤으며, 투여받은 세포는 3 × 105 세포가 더 나은 생존율을 보였습니다. 이러한 결과는 세포와 물질을 성체 제브라피시에 전달하는 IV 주입 방법의 우수한 효능과 신뢰성을 강조합니다.
수용자 캐스퍼 피쉬에서 WKM 세포의 생착 효율은 이식된 세포의 GFP 신호를 모니터링하여 철저히 평가되었습니다. IV 주사를 맞은 물고기에서 GFP 신호는 이식 후 3일째부터 감지될 수 있었으며(그림 2C), 이는 빠르고 효과적인 생착 과정을 나타냅니다. 대조적으로, RO와 심장내 주사를 맞은 물고기는 GFP 신호 출현이 지연되어 생착 속도가 느려졌음을 나타냅니다. 이식 후 17일째까지, 이식된 세포는 주로 신장 골수에 다시 증식했으며, IV를 주입한 물고기에서 가장 뚜렷한 GFP 신호가 관찰되었습니다(그림 2C). 투여군의 KM에 대한 유세포 분석은 IV 주입 방법의 효능을 더욱 강조했습니다. 초기에 3 × 105 세포를 정맥 주사한 수혜자에서 KM의 세포 중 약 18%가 GFP 양성이었으며, 이는 이식을 받지 않은 기증자 Tg(mpo: EGFP) 어류의 신장 골수에서 발견된 비율과 유사한 비율입니다(그림 2D, E). 대조적으로, 심장 내 주사는 약 5%의 GFP 양성 세포를 얻은 반면, RO 방법은 수용자의 신장 골수에서 약 2%의 GFP 양성 세포만 산출했습니다. 또한, 생착 속도는 주입된 세포의 수와 양의 상관관계를 보였는데, 이는 더 많은 세포 용량이 더 큰 생착으로 이어진다는 것을 보여주었습니다. 이러한 결과는 세포 생착 및 신장 골수의 후속 재증식을 촉진하는 IV 주입 기술의 견고성을 확인합니다.
