Purification of HLA-G+ Extravillous Trophoblasts from Human Term Placental Tissues for Phenotyping and Functional Analysis

기대 셀 수율
15 mL의 디시두아 기저 세포에서 기대되는 HLA-G+ EVT는 첫 번째 소화에서 0.1-0.3 × 106 세포, 두 번째 소화에서 0.1-0.6 × 106 세포를 생성한다(총 0.2-0.8 × 106 세포). 30mL의 융모막에서 예상되는 HLA-G+ EVT 수확량은 첫 번째 소화에서 10개의6 세포× 0.4-1.0, 두 번째 소화에서는 0.4-1.5 × 106 세포로 총 0.8-2.5 × 6개의 세포입니다. 분리 내 카스파제 3/7-7-AAD-생 EVT의 비율은 90-95%로 예상됩니다.
1차 HLA-G+ EVT 배양
계통 마커(HLA-G, HLA-C, EGFR)와 세포 사멸 마커의 유동 세포분석 결과, 이 프로토콜이 4일차(분류된 EVT 플레이트 후 96시간)까지 유효한 1차 EVT 배양을 제공함이 확인되었습니다(그림 1B-D). 1차 융모세포 및 기저 굴절성 HDA-G+ EVT의 대표적인 광학 현미경 이미지는 배양 후 24, 48, 72시간 후에 보여줍니다(그림 2A-F).
1차 HLA-G+ EVT와 T 세포의 공동 배양
이 프로토콜은 태반에서 분리된 후 96시간 후 1차 EVT의 생존 가능성을 높였으며, 이는 이전 보고서 1,2,10과 비교해 개선된 수치입니다. 따라서 이는 산모-태아 경계부의 급성 염증을 모방하는 사전 자극 림프구와의 동배양 후 EVT 반응 분석을 가능하게 합니다. EVT는 세포외 유세포 염색을 통해 결정된 다형성 HLA-C 및 면역억제 공동억제 분자인 PD-L1, PD-L2, PVR(CD155로도 알려짐), B7H3, HLA-E를 발현합니다(그림 3A,B). EVT는 HLA-C뿐만 아니라 PD-L1, PD-L2, HLA-E의 발현을 증가시켰으나, 프로토콜 8절에 따르면 활성화된 림프구와 72시간 동안 공동 배양했을 때 PVR과 B7H3의 발현은 증가하지 않았습니다(그림 4A,B). 또한, 7AAD와 카스파제 3/7 염색(그림 4C,D)에서 EVT 사망 증가가 관찰되지 않았습니다.
