1. Set-up
<ol>

	<li>Bereid de werkplek voor door te steriliseren met 70% ethanol. Werk in de buurt van een Bunsenbrander om besmetting van het medium te voorkomen.</li>
	<li>Bij het werken met bacteriën moeten de juiste persoonlijke beschermingsmiddelen en aseptische techniek worden gebruikt. Een laboratoriumjas en handschoenen zijn vereist bij het werken met bacterieculturen.</li>

2. Protocol
<ol>
	<li>Bereiding van Media
	<ol>
		<li>Identificeer het groeimedium waarmee de bacteriën worden gekweekt en bereid zowel vloeibaar bouillon als vast agar (1,5% w/v agar) medium in afzonderlijke autoclaveerbare flessen. Hier werden LB-bouillon en LB-agar bereid voor de groei van Escherichia coli.</li>
		<li>Steriliseer het medium met een semi-vastgedraaide dop in een autoclaaf ingesteld op 121 °C gedurende 35 min.</li>
		<li>Voor agar-medium, na het autoclaveren, plaats in een waterbad ingesteld op 50 °C gedurende 30 minuten om af te koelen. Eenmaal afgekoeld, giet 20-25 mL agar-medium in 100x15mm ronde Petri-schijven. Laat de platen 24 uur op kamertemperatuur stollen voor gebruik.</li>
	</ol>
	</li>
	<li>Initiële voorbereiding van bacteriën
	<ol>
		<li>Van bevroren voorraad, streken bacteriën voor isolatie op geselecteerde media-agar om enkele kolonie-isolaat te verkrijgen. Incuberen in groeiomstandigheden die voor de gekozen bacteriën zijn toegestaan. Hier wordt E. coli op LB-agar gestreken en gedurende 37 °C overnacht (16-18h) geïncubeerd.</li>
		<li>Gebruik een steriele inoculatorlus om een enkele kolonie van de streekplaat te selecteren en inoculeer 4 mL vloeibaar medium in een 15 mL testbuis en groei in de omstandigheden die voor de gekozen bacteriën zijn toegestaan. Hier wordt E. coli gedurende 37 °C geteeld met schudden bij 210 rpm overnacht (16-18h).</li>

	<li>Instelling van de groeicurve

		<li>Voorbereiding van groeiflask
		<ol>
			<li>Autoclaveer geschikte grootte Erlenmeyer kolven. Typisch wordt een 1:5 verhouding van medium tot totale flesvolume gebruikt. Hier wordt 100 mL LB-medium gebruikt in een 500 mL fles.</li>
			<li>Gebruik een serologische pipette om steriele media naar de Erlenmeyer kolf over te brengen.</li>

		<li>Voorbereiding van verdunningsreeks

			<li>Gebruik de overnachtelijke vloeibare cultuur die als onderdeel van stap 2.2.2 is bereid, om de flesmedia te inoculeren met 1:1000 volume van cultuur. Hier wordt 100 μL overnachtelijke vloeibare cultuur toegevoegd aan 100 mL LB-medium.</li>

Latentiefase is de periode die nodig is voor de bacteriën om een fysiologische toestand te bereiken die snel celgroei en deling mogelijk maakt. Deze latentie treedt op omdat het tijd kost voor bacteriën om zich aan hun nieuwe omgeving aan te passen. Zodra de benodigde cellulaire componenten in de latentiefase zijn gegenereerd, gaan bacteriën over naar de exponentiële groeifase waarin DNA-replicatie, RNA-transcriptie en eiwitproductie allemaal met een constante, snelle snelheid plaatsvinden (2). De snelheid van snelle celgroei en deling tijdens de exponentiële fase wordt berekend als de generatietijd, of verdubbelingstijd, en is de snelste snelheid waarmee de bacteriën zich onder de gegeven omstandigheden kunnen repliceren (1). De verdubbelingstijd kan worden gebruikt om verschillende groeiomstandigheden te vergelijken om te bepalen welke gunstiger is voor bacteriële groei. De exponentiële groeifase is de meest reproduceerbare groeitoestand omdat de fysiologie van de bacteriële cellen consistent is in de hele populatie (3). De stationaire fase volgt de exponentiële fase waarin de celgroei plateaut. De stationaire fase wordt veroorzaakt door voedingsstoffenuitputting en/of accumulatie van toxische tussenproducten. Bacteriële cellen blijven in dit stadium overleven, hoewel de snelheid van replicatie en celdeling drastisch is verminderd. De laatste fase is sterven, waarin ernstige voedingsstoffenuitputting leidt tot het lyseren van cellen. Kenmerken van de groeicurve die het meeste informatie bieden, zijn de duur van de latentiefase, de verdubbelingstijd en de maximale celdichtheid die wordt bereikt.
Kwantificering van bacteriën in batchcultuur kan worden bepaald met behulp van zowel kolonievormingseenheden als optische dichtheidsmetingen. Enumeratie door kolonievormingseenheden (CFU) biedt een directe meting van het aantal bacteriële cellen. De standaard eenheid van meting voor CFU is het aantal cultiveerbare bacteriën aanwezig per 1 mL cultuur (CFU/mL) bepaald door seriële verdunning en verspreidingsplaattechnieken. Voor elk tijdstip wordt een 1:10 verdunningsreeks van de batchcultuur uitgevoerd en 100 µl van elke verdunning wordt verspreid geplaat met behulp van een celverspreider.

De platen worden vervolgens overnacht geïncubeerd en klonale kolonies worden geenumereerd. De verdunningsplaat die 30-300 kolonies groeide, wordt gebruikt om de CFU/mL voor het gegeven tijdstip te berekenen (4, 5). Stochastische variatie in kolonietellingen onder de 30 zijn onderhevig aan grotere fouten bij de berekening van CFU/mL en het tellen van kolonies groter dan 300 kan worden onderschat vanwege kolonie-overbevolking en overlapping. Met behulp van de verdunningsfactor voor de gegeven plaat, kan de CFU van de batchcultuur worden berekend voor elk tijdstip.
Optische dichtheid geeft een directe benadering van het aantal bacteriële cellen gemeten met behulp van een spectrofotometer. De optische dichtheid is een maat voor de absorptie van lichtdeeltjes die door 1 cm cultuur gaan en worden gedetecteerd door een fotodiodesensoor (6). De optische dichtheid van een cultuur wordt gemeten in relatie tot een media-leeg en neemt toe naarmate de bacterieële dichtheid toeneemt. Voor bacteriële cellen wordt een golflengte van 600 nm (

Grafieken van kolonievormende eenheden en optische dichtheid zijn twee manieren om groeikinetica te visualiseren. Door de relatie tussen CFU/mL en OD600 te bepalen, geeft de optische dichtheidsgrafiek ook een schatting van CFU/mL in de loop van de tijd. Bacteriën reproduceren zich door middel van een proces dat celdeling wordt genoemd, wat resulteert in twee identieke dochtercellen. Als de groeiomstandigheden gunstig zijn, zullen bacteriepopulaties exponentieel groeien.Bacteriegroeicurven plotten de hoeveelheid bacteriën in een cultuur als functie van de tijd. Een typische groeicurve loopt door vier fasen: latentiefase, exponentiële fase, stationaire fase en doodsfase. De latentiefase is de tijd die nodig is voor bacteriën om een toestand te bereiken waarin ze snel kunnen groeien en delen. Hierna gaan de bacteriën over naar de exponentiële fase, gekenmerkt door snelle celgroei en deling. De snelheid van exponentiële groei van de bacteriecultuur tijdens deze fase kan worden uitgedrukt als de verdubbelingstijd, de snelste snelheid waarmee bacteriën zich onder specifieke omstandigheden kunnen reproduceren. De stationaire fase volgt, waarin de groei van bacteriecellen plateau bereikt en de groei- en sterftepercentages uitgebalanceerd raken vanwege verarming van de omgevingsvoedingsstoffen. Ten slotte gaan de bacteriën de doodsfase in. Dit is waar de bacteriegroei sterk afneemt en ernstige verarming van voedingsstoffen leidt tot het lyseren van cellen.Er zijn twee technieken die kunnen worden gebruikt om de hoeveelheid bacteriën in een cultuur te kwantificeren en een groeicurve te plotten. De eerste hiervan is via kolonievormende eenheden, of CFU's. Om CFU's te verkrijgen, wordt een één-op-tien-serie van negen verdunningen uitgevoerd op regelmatige tijdstippen. De eerste van deze verdunningen, negatief één in dit voorbeeld, bevat 9 mL PBS en 1 mL van de bacteriecultuur. Resulterend in een verdunningsfactor van 1:10. Vervolgens wordt 1 mL van deze oplossing overgebracht naar de volgende buis, negatief twee, resulterend in een verdunningsfactor van 1:100. Dit proces gaat door tot de laatste buis, negatief negen, resulterend in een uiteindelijke verdunningsfactor van 1:1 miljard. Hierna wordt 100 microliter van elke verdunning geplateerd. De platen worden vervolgens geïncubeerd en de klonale kolonies worden geteld. De verdunningsplaat voor een bepaald tijdstip dat tussen de 30 en 300 kolonies groeit, wordt gebruikt om de CFU's per milliliter voor dat tijdstip te berekenen.De tweede veelgebruikte methode voor het meten van bacterieconcentratie is de optische dichtheid. De optische dichtheid van een cultuur kan onmiddellijk worden gemeten, in relatie tot een media-leeg, met een spectrofotometer. Typisch wordt een golflengte van 600 nanometer, ook wel OD600 genoemd, gebruikt voor deze metingen, die toenemen naarmate de celdichtheid toeneemt. Hoewel optische dichtheid minder nauwkeurig is dan CFU's, is het handig omdat het onmiddellijk kan worden verkregen en relatief weinig reagentia vereist. Beide technieken kunnen samen worden gebruikt om een standaardcurve te creëren die de bacterieceltelling van een cultuur nauwkeuriger benadert. In deze video leer je hoe je CFU's en OD600-metingen kunt verkrijgen van getimede seriële verdunningen van E. coli. Laat de platen vervolgens 24 uur op kamertemperatuur stollen.Gebruik om de enkele kolonie-isolaatjes voor te bereiden die later zullen worden gebruikt om een vloeibare bacteriecultuur te produceren, eerder ingevroren voorraad en de juiste strekingstechniek om E. coli voor isolatie op LB-agar te strekken. Incubeer de schaal gedurende een nacht bij 37 graden Celsius. Koel vervolgens een vlam gesteriliseerde inoculatielus af op de agar voordat u een enkele kolonie selecteert van de gestreepte plaat. Inoculeer 4 mL vloeibaar medium in een 15 mL testbuis. Laat vervolgens de E. coli gedurende een nacht bij 37 graden Celsius groeien met schudden bij 210 rpm.Om de 1:1000 volume van bacteriecultuur op te zetten die in de groeicurve zal worden gebruikt, verkrijgt u eerst een autoclaveerbare 500 mL Erlenmeyer-kolf. Gebruik vervolgens een 50 mL serologische pipette om 100 mL steriel medium naar de kolf over te brengen. Vervolgens negen 15 ml testbuizen opeenvolgend labelen als één tot en met negen. Deze nummers komen overeen met de verdunningsfactor die zal worden gebruikt om de kolonievormende eenheid, of CFU, te berekenen. Voeg vervolgens 9 mL 1X PBS toe aan elke buis. Label vervolgens de voorbereide agarplaten met de overeenkomstige tijdstippen en verdunningsfactoren die zullen worden gekweekt. In dit voorbeeld met E. coli, na het begintijdstip, worden tijdstippen genomen om de uur. Gebruik de eerder bereide overnachts vloeibare E. coli-cultuur, inoculeer het medium in de autoclaaf 500 mL Erlenmeyer-kolf met 1:1000 volume van cultuur. Zwaai het medium om de bacteriën gelijkmatig te verdelen.Nadat u een spectrofotometer hebt geblanceerd, maakt u de cuvet schoon met een pluisvrij doek. Verdeel vervolgens 1 mL van de cultuur in de cuvet en plaats deze in de spectrofotometer om de optische dichtheid van de cultuur op tijdstip nul te verkrijgen. Bacterial growth stages and cell counts show how a culture changes over time. This page follows a batch culture as it moves through lag, exponential, stationary, and death phases. It also shows how colony forming units, or CFU, and optical density can be used to measure that growth.

Lag phase is the time bacteria need to adjust to a new environment before fast division begins. During exponential phase, cells grow and divide at the fastest rate, and DNA replication, RNA transcription, and protein production all happen at a steady, rapid pace. The doubling time, also called generation time, describes how long it takes for the population to double. It is useful for comparing which growth conditions are better for a bacterium.

Stationary phase comes after exponential growth slows down and levels off. Nutrient limits and toxic intermediates can cause this plateau. Cells are still alive in this stage, but replication and division are much lower. Death phase follows when severe nutrient depletion leads to cell lysis, or cell breaking apart.

Bacterial numbers in batch culture can be measured by CFU and by optical density. CFU gives a direct count of culturable cells in 1 mL of culture, or CFU/mL. To measure it, the culture is serially diluted 1:10, and 100 µl from each dilution is spread on a plate. After overnight incubation, the plate with 30 to 300 colonies is used to calculate CFU/mL. Counts below 30 can be less reliable, and counts above 300 can be underestimated because colonies crowd together.

Optical density gives a fast estimate of cell density using a spectrophotometer. The reading measures how much light passes through 1 cm of culture and is taken against a media blank. For bacteria, OD600, or optical density at 600 nm, is commonly used. As the culture becomes denser, the OD increases. A standard curve linking OD600 and CFU/mL can turn an instant OD reading into an estimate of bacterial count.

This relationship is most reliable only at lower densities. It can start to break down around 0.3 OD600, and the error becomes greater during stationary and death phases. As cells change shape and release extracellular products into the medium, the OD reading no longer tracks CFU as closely. In the example shown, Escherichia coli is grown in Luria-Bertani, or LB, broth at 37°C for 30 hours. The OD600 curve is measured directly, and values above 1.0 are diluted 1:10, then multiplied by 10 because the spectrophotometer becomes less accurate at high density.

The example E. coli culture shows lag phase to about 1 hour on the OD600 curve and about 2 hours on the CFU/mL curve. Exponential growth runs from about 2 to 7 hours, then the culture plateaus in stationary phase. After about 15 hours, both OD and CFU begin to decline, although the death phase is gradual rather than abrupt. The CFU/mL curve peaks near 2 x 10^9 and falls to about 5 x 10^8 by 30 hours. A linear standard curve between CFU/mL and OD600, based on the earlier time points, lets OD estimate cell density. From these data, the doubling time was calculated as 15 minutes and 19 seconds, which helps compare growth in different media or between different bacteria.
