What is Glycolysis?

- [Spreker] Glycolyse is het stofwisselingsproces om suikers in energie om te zetten. Nadat glucose in het cytoplasma van de cel is doorgedrongen, wordt het in twee fasen omgezet. Het eerste deel kost energie, in de vorm van ATP, om het glucose-molecuul vast te zetten en stapsgewijs te veranderen.

De tweede fase ontrekt energie aan elektronen en benut dit als energierijke NADH- en extra ATP-moleculen . Na meerdere enzymatische stappen zijn er twee triosen, pyruvaat klaar voor transport naar de mitochondriën en opname in de citroenzuurcyclus.

Cellen genereren energie door macromoleculen af te breken. Cellulaire ademhaling is het biochemische proces dat voedselenergie (opgeslagen in de chemische bindingen van macromoleculen) omzet in chemische energie in de vorm van adenosinetrifosfaat (ATP). De eerste stap in dit strak gereguleerde en complexe proces is glycolyse. De term glycolyse komt van het Latijnse glyco (suiker) en lysis (afbraak). Glycolyse heeft twee belangrijke intracellulaire functies: het genereren van ATP en het genereren van tussenliggende metabolieten om in andere routes te voeden. De glycolytische route zet één hexose (een koolhydraat met zes koolstofatomen, zoals glucose) om in twee triosemoleculen (koolhydraat met drie koolstofatomen), zoals pyruvaat, om een netto totaal van twee moleculen ATP te produceren (vier geproduceerd, twee geconsumeerd) en twee moleculen. van nicotinamideadeninedinucleotide (NADH).

<h4>Opheldering van glycolyse</h4>

Halverwege de 19e eeuw stelde Louis Pasteur vast dat micro-organismen glucose afbreken bij afwezigheid van zuurstof (fermentatie). In 1940 werd de volledige glycolyseroute vastgesteld door Gustav Embden, Otto Meyerhof, Jakub Karol Parnas en anderen, waardoor glycolyse nu bekendstaat als de EMP-route.

<h4>Lot van glucose</h4>

Er zijn twee manieren waarop glucose de cel kan binnendringen. Een groep integrale GLUT-eiwitten (glucosetransporter) transporteert glucose naar het cytosol via gefaciliteerde diffusie. Leden van de GLUT-eiwitfamilie zijn aanwezig in specifieke weefsels door het hele menselijk lichaam. Als alternatief verplaatsen transmembraansymportereiwitten glucose tegen de concentratiegradiënt in via secundair actief transport. De symporter gebruikt elektrochemische energie van het pompen van ionen. Voorbeelden zijn de natrium-glucose-gekoppelde transporters in de dunne darm, het hart, de hersenen en de nieren.

Onder aerobe (O2-rijke) en anaerobe (O2-deficiënte) omstandigheden kan glycolyse beginnen zodra glucose het celcytosol binnenkomt. Glycolyse bestaat uit twee hoofdfasen: De eerste fase is een energievereisende voorbereidende stap waarin glucose in de cel wordt vastgehouden en de zes-koolstofstructuur wordt herstructureerd om efficiënt gesplitst te worden. De tweede fase genereert energie en leidt tot de productie van pyruvaat.

<h4>Het lot van Pyruvaat</h4>

Afhankelijk van het zuurstofniveau en de aanwezigheid van mitochondriën kan pyruvaat een van de twee mogelijke lotgevallen hebben. Onder aerobe omstandigheden (O2-aanwezig en mitochondriën aanwezig): Pyruvaat wordt getransporteerd naar de mitochondriën, waar het de citroenzuurcyclus en elektronentransportketen (ETC) ondergaat, resulterend in de productie van CO2, H2O en een grote hoeveelheid ATP. Onder anaerobe omstandigheden (zoals in werkende spieren) of bij afwezigheid van mitochondriën (zoals in prokaryoten): Pyruvaat ondergaat lactaatfermentatie, waarbij het wordt gereduceerd tot lactaat. Interessant is dat gist en sommige bacteriën pyruvaat onder anaerobe omstandigheden kunnen omzetten in ethanol, een proces dat bekend staat als alcoholfermentatie.

<h4>Regulatie van glycolyse</h4>

Een strikte controle en regulering van enzym-gemedieerde metabole routes, zoals glycolyse, is essentieel voor het goed functioneren van een organisme. Controle wordt uitgeoefend door substraatbeperking of enzymgebonden regulatie. Substraatbeperking treedt op wanneer de concentratie van de substraten en producten in de cel bijna in evenwicht is. Bijgevolg bepaalt de beschikbaarheid van het substraat de snelheid van de reactie. Bij enzymgebonden regulatie is de concentratie van de substraten en producten verre van evenwicht. De activiteit van drie enzymen (hexokinase, fosfofructokinase en pyruvaatkinase) bepaalt de reactiesnelheid, die de flux van de totale route regelt.

glycolysis: how cells break down glucose

Glycolysis is the first stage of cellular respiration and a key way cells break down glucose to make energy. Cellular respiration converts food energy from macromolecules into ATP, the cell’s main energy currency. Glycolysis also creates intermediate molecules that can feed into other pathways.

In this pathway, one six-carbon sugar such as glucose is split into two three-carbon molecules, including pyruvate. The process produces a net gain of two ATP molecules, since four ATP are made and two are used. It also generates two molecules of NADH, a carrier used in later energy reactions.

The history of glycolysis began with studies of fermentation. In the mid-1800s, Louis Pasteur showed that microorganisms break down glucose without oxygen. In 1897, Eduard Buchner found that fermentation could still happen in cell-free yeast extracts, which showed that the cytoplasm contains the needed soluble components.

Later, Arthur Harden and William Young found that fermentation slows without inorganic phosphate. They also showed that the process needs both a heat-sensitive part, later linked to enzymes, and a heat-stable part with small molecules such as inorganic ions, ATP, ADP, and coenzymes like NAD. By 1940, Gustav Embden, Otto Meyerhof, Jakub Karol Parnas, and others had established the full pathway, which is now also called the EMP pathway.

Glucose can enter cells in two main ways before glycolysis begins. GLUT proteins move glucose into the cytosol by facilitated diffusion. Transmembrane symporter proteins can also bring glucose in against its concentration gradient by secondary active transport, using energy from ion pumping. Sodium-glucose-linked transporters are examples found in the small intestine, heart, brain, and kidneys.

Glycolysis can begin after glucose enters the cytosol under both aerobic and anaerobic conditions. The pathway has two main phases. The first is an energy-requiring preparatory phase that traps glucose in the cell and rearranges its six-carbon backbone so it can be split efficiently. The second phase releases energy and forms pyruvate.

Pyruvate can follow different paths depending on oxygen and mitochondria. Under aerobic conditions with mitochondria, pyruvate enters the mitochondria and continues through the citric acid cycle and the electron transport chain, where it is oxidized to carbon dioxide, water, and more ATP. Under anaerobic conditions, such as in working muscles, or in cells without mitochondria, such as prokaryotes, pyruvate undergoes lactate fermentation. Yeast and some bacteria can instead convert pyruvate to ethanol through alcohol fermentation.

Glycolysis is tightly regulated because metabolic pathways must be controlled for proper cell function. Regulation can depend on substrate limitation, which occurs when substrate and product levels are near equilibrium and the available substrate sets the reaction rate. It can also depend on enzyme-linked regulation, where the pathway is controlled by key enzymes. Hexokinase, phosphofructokinase, and pyruvate kinase are three enzymes that help determine the overall flux of glycolysis.
