Energy-requiring Steps of Glycolysis

- [Instructeur] Glycolyse begint met een enkele glucosemolecule. Door gebruik van ATP verplaatst het enzyme hexokinase een fosfaatgroep naar de zes koolstof suiker om glucose-6-fosfaat te vormen die opgesloten raakt in de cel door zijn negatieve lading. Vervolgens katalyseert het enzyme fosfogluco-isomerase de omzetting van de fosfoglucose in één van zijn isomeren, fructose-6-fosfaat.

De fosfofructose kan nu gefosforyleerd worden door een snelheidsbeperkd enzyme, fosfofructokinase, om fructose-1,6-difosfaat te vormen. Tenslotte, met de twee fosfaatgroepen gebonden, wordt de suikermolecule gesplitst door aldolase in twee drie koolstof isomeren, glyceraldehyde 3-fosfaat of G3P, en dihydroxyacetonfosfaat, DHAP. Een ander katalyserend enzyme, triosefosfaatisomerase zet de DHAP om in G3P en levert zo twee molecules op.

Dus tijdens deze energieverbruikende fase, worden in totaal netto twee ATP's gebruikt om een initiële glucosemolecule te splitsen in twee kleinere suikers.

Glucose is de bron van bijna alle energie die door organismen wordt gebruikt. De eerste stap in de omzetting van glucose in bruikbare energie wordt glycolyse genoemd. Glycolyse vindt plaats in het cytosol van de cel en verloopt in twee fasen:Energievereisende fase. Energievrijgevende fase. Tijdens de eerste drie stappen wordt glucose omgezet in verschillende vormen en gehecht aan twee fosfaatgroepen, die worden gedoneerd door twee ATP-moleculen, wat resulteert in een onstabiele suiker. In de volgende twee fasen splitst deze onstabiele suiker zich in twee suikerisomeren, die worden omgezet of direct gebruikt in de volgende fase van glycolyse.

<h4>Het proces</h4>

Eerst wordt glucose gefosforyleerd door een fosfaatgroep uit ATP toe te voegen, waardoor het wordt omgezet in glucose-6-fosfaat. Het negatief geladen fosfaat voorkomt dat glucose het hydrofobe celmembraan passeert, waardoor glucose in de cel wordt vastgehouden.

Vervolgens wordt glucose-6-fosfaat omgezet in fructose-6-fosfaat, een isomeer dat nodig is voor de volgende energievereiste stappen van glycolyse.

Daarna wordt fructose-6-fosfaat gefosforyleerd door een tweede ATP-molecuul, wat resulteert in fructose-1,6-bisfosfaat, een onstabiele suiker.

Tot slot splitst deze onstabiele suiker zich in twee suikerisomeren met drie koolstofatomen: Glyceraldehyde-3-fosfaat (G3P. Dihydroxyacetonfosfaat (DHAP). G3P wordt direct gebruikt in de volgende fase van glycolyse, terwijl DHAP wordt omgezet in G3P.

Glycolysis begins in the cytosol, where glucose is prepared for use as energy. This process has two phases: an energy-requiring phase and an energy-releasing phase. The first phase changes glucose into a more reactive, unstable form before it can move into later steps.

In the first step, glucose is phosphorylated, which means a phosphate group is added from ATP. This produces glucose 6-phosphate. The added phosphate is negatively charged, so it cannot cross the hydrophobic cell membrane. As a result, glucose is trapped inside the cell.

Next, glucose 6-phosphate is converted into its isomer, fructose 6-phosphate. An isomer is a molecule with the same atoms but a different arrangement. Fructose 6-phosphate is needed for the remaining energy-requiring steps of glycolysis. It is then phosphorylated by a second ATP molecule, forming fructose 1,6-bisphosphate.

Fructose 1,6-bisphosphate is unstable and splits into two three-carbon sugar isomers. These are glyceraldehyde 3-phosphate and DHAP. Glyceraldehyde 3-phosphate can go straight into the next phase of glycolysis. DHAP is converted into glyceraldehyde 3-phosphate before continuing.
