Pyruvate Oxidation

- [Instructeur] Na glycolyse in eukaryoten komen de geladen pyruvaatmoleculen via actief transport de mitochondriën binnen en ondergaan drie enzymatische reacties. Het proces wordt gekatalyseerd door een complex, het pyruvaatdehydrogenasecomplex, bestaande uit drie enzymen. In de eerste stap wordt de carboxylgroep uit pyruvaat verwijderd en als kooldioxide vrijgemaakt.

Het gestripte molecuul wordt vervolgens geoxideerd tot een acetylgroep en de vrijgekomen elektronen worden vervolgens door NAD plus opgepikt om NADH te vormen. Tot slot wordt de acetylgroep overgebracht naar het co-enzym A, waar het zich aan het zwavelatoom hecht om acetylcenzyme A of acetyl-CoA te creëren, dat in de citroenzuurcyclus terechtkomt om verder te oxideren.

Pyruvate is prepared for the next stage of cellular respiration inside the mitochondria. After glycolysis, the charged pyruvate molecules enter the mitochondria by active transport. There, they go through three enzymatic reactions so the energy stored in pyruvate can be used by the cell.

First, pyruvate dehydrogenase removes the carboxyl group from pyruvate. This step releases carbon dioxide. Next, the stripped molecule is oxidized, and the electrons are picked up by NAD+ to make NADH. The remaining two-carbon fragment becomes acetate.

Finally, coenzyme A attaches to the acetate through its sulfur atom. Coenzyme A is a sulfur-containing compound made from a B vitamin. This forms acetyl coenzyme A, or acetyl CoA. Acetyl CoA then enters the citric acid cycle, where it is oxidized further.
