- [Verteller] In celademhaling, de opbrengst van 30 tot 32 ATP moleculen is het resultaat van de stapsgewijze productie van hoogenergetische-moleculen. Twee ATP's en twee NADH's in de glycolyse, twee NADH's in de oxidatie van pyruvaat, twee ATP's, zes NADH's, en twee FADH2's in de citroenzuurcyclus. En ongeveer 26 of 28 ATP's in oxidatieve fosforylering.

De hoeveelheid ATP is een benadering om drie redenen. Ten eerste, doordat de elektronendragers NADH en FADH2 indirect ATP produceren door waterstofionen in de ruimte tussen de membranen te pompen, creëren ze theoretische fracties van ATP, respectievelijk 2,5 en 1,5 ATP's. Cellulaire ademhaling produceert 30 - 32 ATP per glucosemolecuul. Hoewel het grootste deel van het ATP voortkomt uit oxidatieve fosforylering en de elektronentransportketen (ETC), worden er vooraf 4 ATP gewonnen (2 uit de glycolyse en 2 uit de citroenzuurcyclus).

De ETC is ingebed in het binnenste mitochondriale membraan en bestaat uit vier belangrijke eiwitcomplexen en een ATP-synthase. NADH en FADH2 geven elektronen door aan deze complexen, die protonen in de intermembrane ruimte pompen. Deze verdeling van protonen genereert een concentratiegradiënt over het membraan. De gradiënt drijft de productie van ATP aan wanneer protonen via de ATP-synthase terugstromen naar de mitochondriale matrix.

Voor elke twee elektronen die NADH door complex I passeert, worden in totaal 10 protonen gepompt: complex I en complex III pompen elk 4 protonen, terwijl complex IV 2 protonen pompt. Complex II is niet betrokken bij de elektronenketen die door NADH wordt geïnitieerd. FADH2 geeft echter 2 elektronen door aan complex II, dus per FADH2 worden er in totaal 6 protonen gepompt (4 protonen via complex III en 2 protonen via complex IV).

Er zijn vier protonen nodig om 1 ATP te synthetiseren. Omdat er voor elke NADH 10 protonen worden gepompt, levert 1 NADH 2,5 (10/4) ATP op. Voor elke FADH2 worden zes protonen gepompt, dus 1 FADH2 levert 1,5 (6/4) ATP op.

Cellulaire ademhaling produceert maximaal 10 NADH en 2 FADH2 per glucosemolecuul. Omdat een enkele NADH 2,5 ATP produceert en een enkele FADH2 1,5 ATP produceert, worden 25 ATP + 3 ATP's geproduceerd door oxidatieve fosforylering. Er worden vier ATP geproduceerd vóór oxidatieve fosforylering, wat maximaal 32 ATP per glucosemolecuul oplevert.

Belangrijk is dat glycolyse plaatsvindt in het cytosol en dat de ETC zich in de mitochondriën (in eukaryoten) bevindt. Het mitochondriale membraan is niet permeabel voor NADH. Als gevolg hiervan moeten de elektronen van de 2 NADH die door glycolyse worden geproduceerd, naar de mitochondriën worden getransporteerd. Eenmaal in het mitochondrion kunnen de elektronen worden doorgegeven aan NAD+ of FAD. Gegeven de verschillende ATP-opbrengsten afhankelijk van de elektronendrager, bedraagt de totale opbrengst van cellulaire ademhaling 30 tot 32 ATP per glucosemolecuul.

Cellular respiration makes ATP from glucose in several stages. The process usually produces 30 to 32 ATP per glucose molecule. Most of that ATP comes from oxidative phosphorylation and the electron transport chain, while 4 ATP are made earlier, with 2 from glycolysis and 2 from the citric acid cycle.

The electron transport chain, or ETC, sits in the inner mitochondrial membrane. It has four main protein complexes and ATP synthase, the enzyme that makes ATP. NADH and FADH2 deliver electrons to these complexes. As electrons move through the chain, the complexes pump protons into the intermembrane space and build a gradient across the membrane.

That proton gradient powers ATP production. Protons flow back into the mitochondrial matrix through ATP synthase, and this flow drives ATP formation. For every 2 electrons that NADH sends through complex I, 10 protons are pumped in total. Complex I and complex III each pump 4 protons, and complex IV pumps 2 protons. Complex II is not part of the electron chain started by NADH.

FADH2 follows a different path. It passes 2 electrons to complex II, so a total of 6 protons are pumped per FADH2. In this case, complex III pumps 4 protons and complex IV pumps 2 protons. Four protons are needed to make 1 ATP. That means 1 NADH yields 2.5 ATP, and 1 FADH2 yields 1.5 ATP.

Per glucose molecule, cellular respiration can produce up to 10 NADH and 2 FADH2. Since each NADH makes 2.5 ATP and each FADH2 makes 1.5 ATP, oxidative phosphorylation produces 25 ATP from NADH and 3 ATP from FADH2. Adding the 4 ATP made before that stage gives a maximum of 32 ATP per glucose molecule. In eukaryotes, glycolysis happens in the cytosol, and the ETC is in the mitochondria.

The mitochondrial membrane does not let NADH cross. Because of that, the electrons from the 2 NADH made in glycolysis must be shuttled into the mitochondria. Once inside, those electrons may be passed to NAD+ or FAD. Since the shuttle path can change the carrier used, the total ATP yield of cellular respiration is usually 30 to 32 ATP per glucose molecule.
