DNA-replicatie vindt plaats door nieuwe DNA-strengen te synthetiseren met behulp van bestaande strengen als template. De twee nieuwe dubbele helices bevatten elk een originele template-streng en een nieuw gesynthetiseerde dochterstreng, daarom staat dit proces bekend als semiconservatieve replicatie. Om de replicatie te beginnen, wikkelt een enzym, helicase, de DNA-helix af en verbreekt de waterstofbruggen tussen de twee strengen.Het scheidt vervolgens de afzonderlijke strengen en vormt een Y-vormige structuur, bekend als de replicatievork, waar de template-strengen toegankelijk zijn voor extra enzymen. DNA Replication

DNA-replicatie omvat de scheiding van de twee strengen van de dubbele helix, waarbij elke streng dient als sjabloon waaruit de nieuwe complementaire streng wordt gekopieerd. Na replicatie bevat elk dubbelstrengs DNA één ouderlijke of ‘oude’ streng en één ‘nieuwe’ streng. Dit staat bekend als semiconservatieve replicatie. De resulterende DNA-moleculen hebben dezelfde sequentie en zijn gelijkelijk verdeeld over de twee dochtercellen. Replicatie in prokaryoten DNA-replicatie maakt gebruik van een groot aantal eiwitten en enzymen. Het enzymhelicase scheidt de twee DNA-strengen. Terwijl helicase langs het DNA beweegt, scheidt het de twee strengen en vormt een Y-vormige structuur die bekend staat als de replicatievork. Hierna voegt het enzym primase korte stukjes RNA toe, bekend als primers, om de synthese van DNA door DNA-polymerase, het enzym dat verantwoordelijk is voor de DNA-synthese, te initiëren. Bij bacteriën zijn drie hoofdtypen DNA-polymerasen bekend: DNA pol I, DNA pol II en DNA pol III. DNA pol III is het enzym dat nodig is voor DNA-synthese; DNA pol I en DNA pol II zijn primair nodig voor reparatie. DNA pol III voegt deoxyribonucleotiden, elk complementair aan een nucleotide op de matrijsstreng, één voor één toe aan de 3’-OH-groep van de groeiende DNA-keten. DNA-polymerase III kan zich alleen in de richting van 5’ naar 3’ uitstrekken. De toevoeging van deze nucleotiden kost energie. Deze energie is aanwezig in de bindingen van drie fosfaatgroepen die aan elk nucleotide zijn bevestigd, vergelijkbaar met hoe energie wordt opgeslagen in de fosfaatbindingen van adenosinetrifosfaat (ATP). Wanneer de binding tussen de fosfaten wordt verbroken en difosfaat vrijkomt, maakt de vrijkomende energie de vorming van een covalente fosfodiesterbinding mogelijk door middel van dehydratatiesynthese. De dubbele DNA-helix is antiparallel; dat wil zeggen, de ene streng is georiënteerd in de richting van 5’ naar 3’ en de andere is georiënteerd in de richting van 3’ naar 5’. Tijdens replicatie wordt één streng, die complementair is aan de 3’ tot 5’ ouderlijke DNA-streng, continu gesynthetiseerd in de richting van de replicatievork, omdat polymerase nucleotiden in deze richting kan toevoegen. Deze continu gesynthetiseerde streng staat bekend als de leidende streng. De andere streng, complementair aan het 5'-naar-3'-ouderlijke DNA, groeit weg van de replicatievork, dus het polymerase moet teruggaan naar de replicatievork om te beginnen met het toevoegen van basen aan een nieuwe primer, opnieuw in de richting weg van de replicatievork. Het doet dit totdat het tegen de eerder gesynthetiseerde streng botst, en dan beweegt het weer terug. Deze stappen produceren kleine DNA-sequentiefragmenten die bekend staan als Okazaki-fragmenten, elk gescheiden door RNA-primer. De streng met de Okazaki-fragmenten staat bekend als de achterblijvende streng en de synthese ervan zou discontinu zijn. Na de synthese door DNA-polymerase III worden de primers verwijderd door de exonuclease-activiteit van DNA-polymerase I en worden de gaten opgevuld. De inkepingen die overblijven tussen het nieuw gesynthetiseerde DNA (dat de RNA-primer verving) en het eerder gesynthetiseerde DNA zijn afgesloten door het enzym DNA-ligase dat de vorming katalyseert van een covalente fosfodiesterbinding tussen het 3'-OH-uiteinde van het ene DNA-fragment en het 5'-fosfaatuiteinde van het andere fragment, waardoor de suiker-fosfaatruggengraat van het DNA-molecuul wordt gestabiliseerd. Replicatie in eukaryoten Eukaryotische genomen zijn veel complexer en groter dan prokaryotische genomen en zijn doorgaans samengesteld uit meerdere lineaire chromosomen. Het menselijk genoom heeft bijvoorbeeld 3 miljard basenparen per haploïde set chromosomen, en tijdens de replicatie worden 6 miljard basenparen ingevoegd. Er zijn meerdere replicatieoorsprongen op elk eukaryotisch chromosoom; het menselijk genoom heeft 30.000 tot 50.000 replicatieoorsprongen. De replicatiesnelheid is ongeveer 100 nucleotiden per seconde – 10 keer langzamer dan prokaryotische replicatie. De essentiële stappen van replicatie bij eukaryoten zijn dezelfde als bij prokaryoten. Voordat de replicatie kan beginnen, moet het DNA als sjabloon beschikbaar worden gesteld. Eukaryoot DNA is sterk supercoiled en verpakt, wat mogelijk wordt gemaakt door veel eiwitten, waaronder histonen. Na het initiëren van replicatie, in een proces vergelijkbaar met dat gevonden in prokaryoten, wordt verlenging vergemakkelijkt door eukaryote DNA-polymerasen. De leidende streng wordt continu gesynthetiseerd door het eukaryotische polymerase-enzym pol δ, terwijl de achterblijvende streng wordt gesynthetiseerd door pol ε. Het enzym ribonuclease H (RNase H) verwijdert, in plaats van een DNA-polymerase zoals bij bacteriën, de RNA-primer, die vervolgens wordt vervangen door DNA-nucleotiden. De gaten die overblijven worden afgedicht door DNA-ligase. Net als bij prokaryoten kan het eukaryotische DNA-polymerase alleen nucleotiden toevoegen in de richting van 5’ naar 3’. In de leidende streng gaat de synthese door totdat deze het uiteinde van het chromosoom bereikt of een andere replicatievork in de tegenovergestelde richting. Op de achterblijvende streng wordt DNA in korte stukken gesynthetiseerd, die elk worden geïnitieerd door een afzonderlijke primer. Wanneer de replicatievork het einde van het lineaire chromosoom bereikt, wordt de Er is geen plek om een primer te maken voor het DNA-fragment dat aan het uiteinde van het chromosoom moet worden gekopieerd. Deze uiteinden blijven dus ongepaard en kunnen na verloop van tijd steeds korter worden naarmate de cellen zich blijven delen. DNA replication copies genetic material before a cell divides. Each original DNA strand serves as a template for a new complementary strand. The result is semiconservative replication, which means each new DNA molecule contains one old strand and one new strand. The two daughter cells then receive DNA with the same sequence.

Replication starts when helicase separates the two strands of the double helix. This creates a Y-shaped replication fork. Primase then adds short RNA primers that give DNA polymerase a place to begin DNA synthesis. In bacteria, the main DNA polymerases are DNA pol I, DNA pol II, and DNA pol III. DNA pol III carries out DNA synthesis, while DNA pol I and DNA pol II mainly help with repair.

DNA pol III adds deoxyribonucleotides one at a time to the 3'-OH end of the growing strand. It can extend DNA only in the 5' to 3' direction. The energy for this process comes from the three phosphate groups on each nucleotide. When a phosphate bond is broken and diphosphate is released, that energy helps form the phosphodiester bond in the DNA backbone.

Because the DNA double helix is antiparallel, the two template strands face opposite directions. The strand copied toward the replication fork is made continuously and is called the leading strand. The other strand is copied away from the fork. It must start over with new primers as the fork opens, so it is made in short stretches called Okazaki fragments. This strand is the lagging strand, and its synthesis is discontinuous.

After DNA pol III finishes, DNA pol I removes the RNA primers in bacteria, and the gaps are filled in. DNA ligase then seals the remaining nicks by forming covalent phosphodiester bonds between adjacent DNA fragments. This step strengthens the sugar-phosphate backbone of the new DNA molecule.

Eukaryotic DNA replication follows the same basic steps, but the genome is larger and more complex. Eukaryotic chromosomes are linear and have many origins of replication. In humans, the genome has 3 billion base pairs per haploid set, and about 6 billion base pairs are copied during replication. Human chromosomes also have about 30,000 to 50,000 origins of replication, and replication occurs at about 100 nucleotides per second, which is much slower than in prokaryotes.

Before replication begins, eukaryotic DNA must be made available as a template. It is tightly packed and supercoiled, and histones help organize this packaging. After initiation, eukaryotic DNA polymerases carry out elongation. Polymerase delta (pol δ) synthesizes the leading strand, and polymerase epsilon (pol ε) synthesizes the lagging strand. RNase H removes the RNA primers, DNA nucleotides replace them, and DNA ligase seals the final gaps. Because eukaryotic DNA polymerase also works only in the 5' to 3' direction, the lagging strand is still made in short pieces. Near the ends of linear chromosomes, there is no place to add a final primer, so these ends remain unpaired and can become shorter over time as cells keep dividing.
