[Verteller] Nucle&#xEF;nezuren zijn polymeren van nucleotiden, monomeren bestaand uit een pentosesuiker, een fosfaatgroep en een stikstofbase.
 Binnen de suikermolecule zijn de vijf koolstoffen genummerd van &#xE9;&#xE9;n tot vijf accent waarbij elke notatie vertelt welke groep er aan bindt.
 Bijvoorbeeld, de tweede koolstof kan gebonden worden aan een waterstof item, waardoor een deoxyribosesuiker zoals in desoxyribonucle&#xEF;nezuur of DNA.
Anderzijds kan een hydroxylgroep die plaats innemen om een ribose suiker te vormen, zoals in ribonucle&#xEF;nezuur of RNA.
 Om de afwisselende fosfaatsuiker ruggengraat te vormen, wordt de vijf accent koolstof gebonden aan de fosfaatgroep.
 Dit bindt aan de drie accent koolstof van de volgende nucleotide, waardoor een keten ontstaat via fosfodi-ester binding.
Deze ordening bepaalt de richting waarin de sequentie van nucle&#xEF;nezuren wordt gelezen, vijf accent naar drie accent op een enkele streng.
 De laatste belangrijke koolstof, &#xE9;&#xE9;n accent, wordt gebonden aan een stikstofbase met enkele of dubbele ringstructuur.
 Pyrimidines, zoals cytosine, thymine en uracil hebben een enkele organische ring, waar purines, adenine en guanine, twee ringen hebben.
Waterstofbruggen tussen stikstofbases van twee afzonderlijke strengen vormen de dubbele helix van DNA.
 Guanine bindt altijd aan cytosine en adenine bindt met thymine.
 RNA bevat uracil in plaats van thymine en blijft een enkele streng.

Nucle&#xEF;nezuren zijn de belangrijkste macromoleculen voor de continu&#xEF;teit van het leven. Ze dragen de genetische blauwdruk van de cel en bevatten instructies voor de werking ervan.

DNA en RNA

De twee belangrijkste soorten nucle&#xEF;nezuren zijn deoxyribonucle&#xEF;nezuur (DNA) en ribonucle&#xEF;nezuur (RNA). DNA is het genetische materiaal in alle levende organismen, vari&#xEB;rend van eencellige bacteri&#xEB;n tot meercellige zoogdieren. Het bevindt zich in de kern van eukaryoten en in de organellen, chloroplasten en mitochondri&#xEB;n. In prokaryoten is het DNA niet omgeven door een membraan.

Het gehele genetische materiaal van de cel wordt het genoom genoemd, en de studie van genomen is genomica. In eukaryote cellen, maar niet in prokaryoten, vormt DNA een complex met histon-eiwitten om chromatine te vormen, de substantie van eukaryote chromosomen. Een chromosoom kan tienduizenden genen bevatten. Veel genen bevatten de informatie om eiwitten te maken. Andere genen coderen voor RNA-producten. DNA regelt alle cellulaire activiteiten door de genen &#x22;aan&#x22; of &#x22;uit&#x22; te zetten.

De andere soort nucle&#xEF;nezuur, RNA, is voornamelijk betrokken bij eiwitsynthese. De DNA-moleculen verlaten de kern nooit, maar gebruiken in plaats daarvan een tussenpersoon om te communiceren met de rest van de cel. Deze tussenpersoon is het boodschapper-RNA (mRNA). Andere soorten RNA&#x2014;zoals rRNA, tRNA en microRNA&#x2014;zijn betrokken bij eiwitsynthese en de regulatie daarvan.

DNA en RNA bestaan uit monomeren die nucleotiden worden genoemd. Elke nucleotide bestaat uit drie componenten: een stikstofbase, een pentose (vijf-koolstof) suiker, en een fosfaatgroep. Elke stikstofbase in een nucleotide is gebonden aan een suikermolecuul, dat weer is gebonden aan een of meer fosfaatgroepen. De stikstofbasen, belangrijke componenten van nucleotiden, zijn organische moleculen die zo genoemd worden omdat ze koolstof en stikstof bevatten. Ze worden basen genoemd omdat ze een amino-groep bevatten die het potentieel heeft om een extra waterstof te binden, waardoor de waterstofionenconcentratie in de omgeving wordt verlaagd en het daardoor basischer wordt.

Elk nucleotide in DNA bevat een van de vier mogelijke stikstofbasen: adenine (A), guanine (G), cytosine (C), en thymine (T). Adenine en guanine worden purines genoemd. De primaire structuur van purines bestaat uit twee koolstof-stikstofringen. Cytosine, thymine en uracil worden pyrimidines genoemd, die een enkele koolstof-stikstofring hebben als hun primaire structuur. Elke van deze basale koolstof-stikstofringen heeft verschillende functionele groepen eraan vast. In de moleculaire biologie wordt shorthand de stikstofbasen aangeduid met hun symbolen A, T, G, C, en U. DNA bevat A, T, G en C; terwijl RNA A, U, G en C bevat.

De pentosesuiker in DNA is deoxyribose, en in RNA is de suiker ribose. Het verschil tussen de suikers is de aanwezigheid van de hydroxylgroep op het tweede koolstofatoom van ribose en waterstof op het tweede koolstofatoom van deoxyribose. Het fosfaatresidu hecht zich aan de hydroxylgroep van de 5&#x2032;-koolstof van de ene suiker en de hydroxylgroep van de 3&#x2032;-koolstof van de suiker van het volgende nucleotide, waardoor een 5&#x2032;&#x2013;3&#x2032; fosfodiesterbinding ontstaat.

DNA dubbele helixstructuur

DNA heeft een dubbele helixstructuur. De suiker- en fosfaatgroepen liggen aan de buitenkant van de helix en vormen de ruggengraat van het DNA. De stikstofbasen zijn in het binnenste gestapeld, zoals een paar traptreden. Waterstofbruggen binden de basen met elkaar. Elk basenpaar in de dubbele helix is gescheiden van het volgende basenpaar door 0,34 nm. De twee strengen van de helix lopen in tegengestelde richtingen, wat betekent dat het 5&#x2032;-koolstofuiteinde van de ene streng tegenover het 3&#x2032;-koolstofuiteinde van de overeenkomstige streng zal liggen. Alleen bepaalde soorten basenparing zijn toegestaan&#x2014;A kan zich binden met T, en G kan zich binden met C. Dit is de complementaire base-regel. Met andere woorden, de DNA-strengen zijn complementair aan elkaar.

RNA

Ribonucle&#xEF;nezuur (RNA) is voornamelijk betrokken bij het proces van eiwitsynthese onder de leiding van DNA. RNA is meestal enkelstrengig en bestaat uit ribonucleotiden die met fosfodi&#xEB;sterbindingen aan elkaar zijn gekoppeld.

Er zijn vier belangrijke soorten RNA: boodschapper-RNA (mRNA), ribosomaal RNA (rRNA), transport-RNA (tRNA), en microRNA (miRNA). De eerste, mRNA, draagt het bericht van DNA, dat alle cellulaire activiteiten in een cel regelt. Als een cel een bepaald eiwit nodig heeft, wordt het gen daarvoor &#x22;aan&#x22; gezet en wordt het boodschapper-RNA in de kern gesynthetiseerd. De RNA-basensequentie is complementair aan de coderingssequentie van het DNA waarvan het is gekopieerd. In het cytoplasma interageert het mRNA met ribosomen en andere cellulaire machines.

Het mRNA wordt in sets van drie basen gelezen, bekend als codons. Elk codon codeert voor &#xE9;&#xE9;n enkel aminozuur. Op deze manier wordt het mRNA gelezen en het eiwitproduct gemaakt. Ribosomaal RNA (rRNA) is een belangrijke component van ribosomen waaraan het mRNA bindt. Het rRNA zorgt voor de juiste uitlijning van het mRNA en de ribosomen. Het rRNA van het ribosoom heeft ook een enzymatische activiteit (peptidyl transferase) en katalyseert de vorming van peptidebindingen tussen twee uitgelijnde aminozuren. Transport-RNA (tRNA) is een van de kleinste van de vier soorten RNA, meestal 70&#x2013;90 nucleotiden lang. Het transporteert het juiste aminozuur naar de eiwitsyntheseplaats. Het is de basenparing tussen het tRNA en mRNA die ervoor zorgt dat het juiste aminozuur zich in de polypeptideketen voegt. MicroRNA&#x27;s zijn de kleinste RNA-moleculen, en hun rol is het reguleren van genexpressie door in te grijpen in de expressie van bepaalde mRNA-berichten.

Hoewel RNA enkelstrengig is, vertonen de meeste RNA-soorten uitgebreide intramoleculaire basenparen tussen complementaire sequenties, wat een voorspelbare driedimensionale structuur cre&#xEB;ert die essentieel is voor hun functie.
