Introduction to Enzymes

Enzymes are biological catalysts that accelerate reaction rates without being consumed.&#160; They are typically proteins located in the cytoplasm, inside organelles, and in cell or organelle membranes.
Enzymes may function intracellularly or can be secreted extracellularly. Most enzymes require a specific temperature and pH to operate at peak efficiency.
Substrates are the reactants that specifically bind to active sites, small regions on the enzyme where the reaction occurs. The remaining parts of the enzyme provide structure to the enzyme or interact with other molecules to promote or inhibit the reaction.
Enzymes change their shape to bind their substrates and catalyze the conversion of substrates into products.&#160; After the reaction, the enzyme releases the product and returns to its original conformation, which allows the enzyme to catalyze additional rounds of the reaction.
Some enzymes only function when associated with cofactors, which assist an enzyme-catalyzed reaction. Non-protein molecules such as vitamins, metal ions, or ATP act as cofactors for various enzymes.

De meeste enzymen zijn eiwitten die biochemische reacties versnellen zonder te worden geconsumeerd. Enzymen bevatten een of meer actieve sites die de substraten binden en omzetten in producten. Veel enzymen hebben ook allosterische plaatsen, extra plaatsen waar andere moleculen dan substraten binden om het enzym te activeren of te remmen.

Sommige enzymen vereisen de aanwezigheid van cofactoren – metaalionen of organische moleculen voor katalyse. Cofactoren binden zich aan de actieve plaats van een enzym en helpen bij de omzetting van substraten in producten. Een enzym zonder cofactor is een apo-enzym en wordt een holo-enzym zodra het een cofactor bindt. Een cofactor die een organisch molecuul is, wordt een co-enzym genoemd. Remmers zijn moleculen die een door enzym gekatalyseerde reactie tegenhouden. Competitieve remmers lijken op substraatmoleculen en concurreren ermee om zich aan de actieve plaatsen te binden. Daarentegen binden niet-competitieve remmers zich aan allosterische plaatsen en veranderen de enzymconformatie om substraatbinding te verminderen.

Enzymes are proteins that speed up biochemical reactions without being used up. Humans have used enzymes since about 7000 BCE, when they fermented sugars to make alcohol without knowing the reaction was enzyme-catalyzed. The word enzyme was coined by Wilhelm Kuhne in 1877. It comes from the Greek words en, meaning “in” or “within,” and zyme, meaning “yeast.”

Enzymes work through active sites, which are regions that bind substrates and help convert them into products. Many enzymes also have allosteric sites. These are extra binding sites where molecules other than substrates can attach and either activate or inhibit the enzyme.

Some enzymes need cofactors to carry out catalysis. Cofactors are metal ions or organic molecules that bind to the active site and help the reaction happen. An enzyme without its cofactor is called an apoenzyme. When the cofactor is attached, it becomes a holoenzyme. An organic cofactor is called a coenzyme, and most coenzymes are derived from vitamins.

Enzyme activity can also be blocked by inhibitors. Competitive inhibitors are similar to the substrate and compete for the active site. Non-competitive inhibitors bind to allosteric sites instead. This changes the enzyme’s shape and reduces substrate binding.
