ATP and Macromolecule Synthesis

Macromolecules such as proteins, polynucleotides, carbohydrates, and lipids are polymers of amino acids, nucleotides, monosaccharides, and fatty acids.
Since the synthesis of these macromolecules is energetically unfavorable, the energy released by ATP hydrolysis, a favorable exergonic process, is used to power these reactions.
For example, during polynucleotide chain synthesis, the terminal phosphates from two ATP molecules are released by hydrolysis. These phosphates are then transferred to nucleoside monophosphate, converting it into a high-energy intermediate called nucleoside triphosphate.
This intermediate attaches to the growing end of the polynucleotide chain by releasing pyrophosphate.
Synthesis of polymers can be oriented in two ways. In the case of lipids and proteins, synthesis occurs by head polymerization, where the reactive bond required for the condensation reaction is carried to the end of the growing polymer. Each monomer brings the reactive bond, required for the addition of the following monomer.
In tail polymerization, as seen in the synthesis of polynucleotides and carbohydrates, the reactive bond is carried by the incoming monomer, and is used immediately for its own addition.

Biologische macromoleculen zijn organische verbindingen, voornamelijk samengesteld uit koolstofatomen. De koolstofatomen zijn covalent gebonden met waterstof, zuurstof, stikstof en andere kleine elementen. Er zijn vier belangrijke klassen van biologische macromoleculen: koolhydraten, lipiden, eiwitten en nucleïnezuren.

De meeste macromoleculen zijn samengesteld uit afzonderlijke subeenheden of bouwstenen, monomeren genoemd. De monomeren combineren met elkaar met behulp van covalente bindingen om grotere moleculen te vormen die bekend staan als polymeren.

Het omzetten van monomeren in polymeren is een energetisch intensief proces. Daarom vertrouwen cellen op ATP, de energievaluta van de cel, om deze biochemische processen aan te drijven. Tijdens de omzetting van glucose in glycogeen wordt ATP bijvoorbeeld gehydrolyseerd tot ADP en anorganisch fosfaat (Pi), waardoor vrije energie vrijkomt. Het vrijgekomen anorganische fosfaat bindt zich aan glucose en zet dit om in glucose-6-fosfaat dat nodig is voor de glycogeensynthese.

Deze cyclus van ATP wordt voortdurend afgebroken in ADP-herhalingen om alle levensprocessen aan te drijven. Net als een oplaadbare batterij wordt ADP continu geregenereerd tot ATP door de herbevestiging van een derde fosfaatgroep.

ATP helps cells build biological macromolecules. These organic compounds are mainly made of carbon atoms bonded to hydrogen, oxygen, nitrogen, and small amounts of other elements. The four major classes of biological macromolecules are carbohydrates, lipids, proteins, and nucleic acids.

Most macromolecules start as small subunits called monomers. Cells join monomers with covalent bonds to make larger molecules called polymers. This building process needs energy, so cells depend on ATP, the energy currency of the cell, to drive these biochemical reactions.

One example is the conversion of glucose into glycogen. During this process, ATP is hydrolyzed into ADP and inorganic phosphate, or Pi, and releases free energy. The inorganic phosphate then binds to glucose and forms glucose 6-phosphate, which is needed for glycogen synthesis.

ATP use does not stop there. The cycle repeats as ATP breaks down to ADP and is then regenerated when a third phosphate group is added back. Like a rechargeable battery, this ATP and ADP cycle supports many of life's processes.
