Nomenclature of Aryl and Heterocyclic Amines

Phenylamine&#8212;the simplest aryl amine, and its derivatives are known by their common names, with prefixes o-, m- , and p- used to indicate the point of substitution.
Systematic nomenclature refers to them as derivatives of aniline. The ring is numbered from the carbon atom bearing the &#8211;NH2 group and continues in the direction to give the lowest locant to the substituent.
For more substituted aryl amines, the first point of difference is assigned the lowest locant, and the substituents are listed alphabetically.
Secondary and tertiary aryl amines are named N-substituted anilines.
Saturated and unsaturated heterocyclic amines have specific names based on the ring size and the heteroatoms present.
The numbering starts at the N atom or the heteroatom with the highest atomic weight and continues in the direction to give the lowest locant to the other heteroatom.
Alternatively, prefixes aza- or diaza- along with the locants, indicate the number of N atoms in the ring.

 Voor de systematische nomenclatuur wordt de ring genummerd vanaf het stikstofhoudende koolstofatoom in de richting die de laagste locant voor de substituent oplevert. De volgorde van nummering is om het laagste getal te krijgen voor het eerste verschilpunt in het geval van aminen met meer dan één substituent op de ring. Zoals geïllustreerd in figuur 2 zijn de substituenten alfabetisch gerangschikt. Als alternatief worden ze ook wel derivaten van benzeenamine genoemd.

 Secundaire en tertiaire aromatische aminen zijn vernoemd naar de oorspronkelijke primaire aminen, waarbij N-substituties voorafgaand aan de oorspronkelijke naam van het amine, zoals weergegeven in Figuur 3.

 Verzadigde en onverzadigde heterocyclische aminen, waarbij N deel uitmaakt van de heterocyclus, hebben specifieke namen gebaseerd op de ringgrootte en heteroatomen. Figuur 4 toont enkele biologische aminen die bekend staan onder hun gebruikelijke namen: pyrrolidine, pyrrool, piperidine, pyridine en imidazool.

 Voor de nomenclatuur van heterocyclische aminen begint de nummering bij het heteroatoom met een hoger atoomgewicht voor aminen die naast N ook andere heteroatomen bevatten. Zoals bijvoorbeeld geïllustreerd in figuur 5, wordt de zwavel in thiazool genummerd één en gaat verder in de richting om de laagste nummering te geven ten opzichte van het andere heteroatoom, stikstof. Een soortgelijke waarneming wordt opgemerkt bij de aanwezigheid van zuurstof in morfoline.

 Als alternatief worden de voorvoegsels aza- of diaza-, samen met het locantnummer om het aantal koolstofatomen aan te geven dat is vervangen door het stikstofatoom, gebruikt vóór het hoofdalkaan. Dit komt vaker voor in de nomenclatuur van een bicyclisch systeem wanneer het N-atoom deel uitmaakt van een gefuseerd ringsysteem, zoals in DBU en DBN, zoals weergegeven in figuur 6.

Naming aniline and ring amine derivatives follows clear rules for aromatic and heterocyclic amines. The simplest aromatic amine is phenylamine, which has an –NH2 group directly attached to an aromatic ring. The common name for this skeleton is aniline.

Functionalized anilines often use the prefixes ortho-, meta-, and para- to show where a substituent is placed on the ring. Many of these compounds also have trivial, or common, names. For systematic naming, the ring is numbered from the carbon attached to nitrogen. Numbering then continues in the direction that gives the lowest locant to the next substituent, and substituents are listed in alphabetical order. These compounds may also be named as benzenamine derivatives.

Secondary and tertiary aromatic amines are named from the parent primary amine. In these names, N-substituents are written before the parent amine name. This shows which groups are attached to the nitrogen atom itself.

Heterocyclic amines also have specific names. These compounds contain nitrogen as part of the ring, and they may be saturated or unsaturated. Common biological examples include pyrrolidine, pyrrole, piperidine, pyridine, and imidazole.

When a heterocycle contains more than one heteroatom, numbering starts at the heteroatom with the higher atomic weight. The numbering then continues in the direction that gives the other heteroatom the lowest possible number. For example, sulfur is numbered first in thiazole, and oxygen is numbered first in morpholine.

Another naming style uses the prefixes aza- or diaza-. These prefixes show how many carbons in the parent alkane are replaced by nitrogen atoms, and the locant number gives the nitrogen position. This approach is often used for bicyclic fused ring systems, such as DBU and DBN.
