The proton NMR spectrum of an amine shows the N&#8211;H signal as a broad band in the region of 0.5 to 4 ppm.
The peak&#39;s position depends on the extent of hydrogen bonding, the solvent&#39;s nature, the amine&#39;s concentration, and the temperature conditions.
Because the N&#8211;H protons are easily exchangeable, no coupling occurs with the neighboring protons&#8212;the peak is broad, unsplit, and difficult to identify.
A D2O&#160;shake exchanges the N&#8211;H protons for the N&#8211;D deuterons causing the N&#8211;H peak to disappear, confirming the presence of amine protons in the sample.
Since the &#945; carbon protons are deshielded by nitrogen, they have higher chemical shift values than the &#946; carbon protons that are far less deshielded and appear further upfield.
In the 13C NMR spectrum, the &#945; carbon shows the highest chemical shift as the nitrogen atom greatly deshields it.
An increasing distance between carbon and nitrogen decreases the deshielding effect, causing the carbon atom&#39;s chemical shift value to decrease.&#160;

Omdat amineprotonen in oplossing een snelle protonenuitwisseling ondergaan, zijn de protonen labiel en nemen daarom niet deel aan enige splitsing met aangrenzende protonen. De waargenomen piek is dus breed en geeft geen enkele informatie over de aangrenzende protonenomgeving. Dit probleem wordt opgelost en NH-protonen worden identificeerbaar gemaakt door D_2O aan het mengsel toe te voegen. De toevoeging zorgt ervoor dat de NH-protonen uitwisselen met deuteronen, wat leidt tot het verdwijnen van NH-protonpieken. Deze verdwijning is indicatief voor de aanwezigheid van labiele protonen in het monster.


 Bij alifatische aminen worden de α-protonen afgeschermd door het elektronenopnemende stikstofatoom. Hierdoor vertonen de α-protonen hogere chemische verschuivingen (δ 2,2 tot 2,9 ppm) dan β-protonen (δ 1 tot 1,7 ppm) die minder afgeschermd zijn vanwege de grotere afstand tot het stikstofatoom.


 In de ^13C NMR-spectroscopie vertonen de α-koolstofatomen van alifatische aminen de hoogste chemische verschuivingswaarden variërend van δ 30 tot 60 ppm vanwege het onschadelijke effect van elektronegatieve stikstof.

Amines in NMR spectroscopy show clear signal patterns that help identify their structure. In proton NMR, primary amines and secondary amines give a broad N–H signal between δ 0.5 and 5 ppm. The exact position can change with sample concentration, hydrogen bonding, and the solvent used.

The N–H protons in amines are labile, which means they exchange quickly in solution. Because of this fast proton exchange, they do not split with nearby protons. Their peak stays broad and does not reveal the environment of adjacent protons.

A D2O test can make these N–H protons easier to identify. When deuterium oxide is added, the N–H protons exchange with deuterons and the N–H peaks disappear. This loss of signal shows that labile protons are present in the sample.

Aliphatic amines also shift nearby carbon and hydrogen signals. The α protons, which are next to the nitrogen atom, are deshielded and appear at higher chemical shifts, about δ 2.2 to 2.9 ppm. The β protons are farther away and appear at lower shifts, about δ 1 to 1.7 ppm. In 13C NMR spectroscopy, the α carbons of aliphatic amines give the highest chemical shift values, from δ 30 to 60 ppm, because nitrogen pulls electron density away.
